KEYWORDSNovel miniaturized polyvinyl chloride membrane sensors in all-solid state graphite and platinum wire supports were developed, electrochemically evaluated and used for the assay of naftidrofuryl oxalate. The naftidrofuryl oxalate sensors were based on the formation of an ionassociation complex between the drug cation and tetrakis-(4-chlorophenyl)borateanionic exchanger as electroactive material and sulfocalix-8-arene as ionophore dispersed in a polyvinyl chloride matrix. Linear responses of 10 -2 -10 -6 M and 10 -2 -10 -5 M with cationic slopes of 56.9 mV and 54.1 mV over the pH range 2-5 were obtained by using the naftidrofuryl oxalate-coated graphite (Sensor 1) and platinum wire (Sensor 2) membrane sensors, respectively. The utility of 4-sulfocalix-8-arene as ionophore had a significant influence on increasing the membrane sensitivity and selectivity. The methods were also used to determine the intact drug in the presence of it's degradate in Praxilene ® tablets, plasma and cerebrospinal fluid with good recovery. Sensor 1 was used to study the kinetics of naftidrofuryl oxalate alkaline degradation that was found to follow a pseudo first-order reaction. The activation energy could be estimated from the Arrhenius plot to be 12.572 Kcal/mol. L-Dopa is the intermediate precursor of the neurotransmitter dopamine. Unlike dopamine, L-dopa easily enters the central nervous system. L-Dopa, as one of the catecholamines, is widely used as a source of dopamine in the treatment of most patients with Parkinson's disease and epilepsy. Graphene (GR) is ideally suited for implementation in electrochemical applications due to its reported large electrical conductivity, large surface area, unique heterogeneous electron transfer rate, and low production costs. This work reports the synthesis of GR using a modified Brodie method and its application for the electrochemical determination of L-dopa in real samples. Electrochemical measurements were performed at glassy carbon electrode modified with graphene (GR/GCE) via drop casting method. Cyclic voltammograms of L-dopa at GR/GCE showed an increased current intensity compared with GCE. All the measurements were done in phosphate buffer solution 0.1 M (pH 6.2) and the oxidation peak was observed at 0.27 V vs. Ag/AgCl. The effect of scan rate showed that oxidation of L-dopa on GR/GCE was surface controlled. The oxidation peak current of L-dopa gradually increased with increasing accumulation time from 0 to 300 s and accumulation potential from 0.0 to 0.3 V and reached the maximum current response at 240 s and 0.2 V for the accumulation time and accumulation potential, respectively. Voltammetric peak currents showed a linear response for L-dopa concentration in the range of 0.04 to 79 μM and a detection limit of 0.022 μM (22 nM).The relative standard deviation for five determinations of 50 μM L-dopa was 0.52 %.

Three sensitive, selective and precise stability-indicating methods for the determination of the anti-Alzheimer's drug, rivastigmine hydrogen tartrate (RIV) in the presence of its alkaline degradation product (major metabolite, NAP 226-90) and in pharmaceutical formulation were developed and validated. The first method is a second derivative (D(2)) spectrophotometric one, which allows the determination of RIV in the presence of its degradate at 262 nm (corresponding to zero crossing of the degradate) over a concentration range of 50-500 microg/ml with mean percentage recovery 100.18 +/- 0.628. The second method is the first derivative of the ratio spectra (DD(1)) by measuring the peak amplitude at 272 nm over the same concentration range as (D(2)) spectrophotometric method, with mean percentage recovery 99.97 +/- 0.641. The third method is a TLC-densitometric one, where RIV was separated from its degradate on silica gel plates using methanol:butanol:H(2)O:ammonia (5:4:1:0.01 v:v:v) as a developing system. This method depends on the quantitative densitometric evaluation of thin layer chromatogram of RIV at 263 nm over a concentration range of 20-160 microg/spot, with mean percentage recovery 100.19 +/- 1.344. The selectivity of the proposed methods was tested using laboratory-prepared mixtures. The proposed methods have been successfully applied to the analysis of RIV in pharmaceutical dosage forms without interference from other dosage form additives and the results were statistically compared with reference method.
