Mimotope discovery as a tool to design a vaccine against Zika and dengue viruses

Abstract: Vaccine development against dengue virus is challenging because of the antibody‐dependent enhancement of infection (ADE), which causes severe disease. Consecutive infections by Zika (ZIKV) and/or dengue viruses (DENV), or vaccination can predispose to ADE. Current vaccines and vaccine candidates contain the complete envelope viral protein, with epitopes that can raise antibodies causing ADE. We used the envelope dimer epitope (EDE), which induces neutralizing antibodies that do not elicit ADE, to design a vacc… Show more

“…The current detection methods are costly, and synthetic methods of gizzerosine are scarce. ,, Moreover, the small size of the gizzerosine, and the presence of a rare secondary amine within the molecule complicate its labeling. ,, Recently, immunoassay technology based on anti-idiotype antibodies that can specifically bind to the antibody via imitating the epitope of the antigen and compete with the binding site of the analyte are used in immunodiagnosis, , pesticide residue detection, and vaccine development. , The marker of this assay is an easily labeled antibody, which makes the assay independent of hapten availability . The immunization of BALB/c mice with anti-gizzerosine mAb 3H4 conjugated to BC prepares 34 potential anti-idiotype antibodies.…”

“…In the area of vaccine development, Cuevas-Juárez et al (2023) applied phage display technology to produce epitopes (or mimotopes) that when attached to AAV virus-like particles (VLP) can produce an antibody response to recognize and potentially protect against zika and dengue viruses. In another study, Lorenzo et al (2023) described a process to purify HIV-1 Gag VLPs from contaminating host EVs using a series of downstream isolation stages including multiple filtration and chromatography steps.…”
