Mimicking Elementary Reactions of Manganese Lipoxygenase Using Mn-hydroxo and Mn-alkylperoxo Complexes

Abstract: Manganese lipoxygenase (MnLOX) is an enzyme that converts polyunsaturated fatty acids to alkyl hydroperoxides. In proposed mechanisms for this enzyme, the transfer of a hydrogen atom from a substrate C-H bond to an active-site MnIII-hydroxo center initiates substrate oxidation. In some proposed mechanisms, the active-site MnIII-hydroxo complex is regenerated by the reaction of a MnIII-alkylperoxo intermediate with water by a ligand substitution reaction. In a recent study, we described a pair of MnIII-hydroxo … Show more

“…A linear fit of k obs versus TEMPOH concentration (Figure 12, left) yielded a second-order rate constant for the reaction of TEMPOH with 2 of 6.1 M −1 s −1 at 298 K in CH 3 CN. This rate fits well with those of similar Mn III -hydroxo complexes, including [Mn III (OH)(dpaq 5NO2 )] + (7 M −1 s −1 ) [52] and [Mn III (OH)( 6Me dpaq)] + (3.4 M −1 s −1 ) [54].…”
