Midkine induces the transformation of NIH3T3 cells

Abstract: Summary Midkine (MK) is a heparin-binding growth factor and is frequently expressed at high levels in many human carcinomas. To investigate further the roles of MK in the regulation of cell growth, we introduced MK expression in NIH3T3 cells. A mixture of transfectants of an MK expression vector, but not a control vector, formed colonies in soft agar, showed an elevated cell number at confluence, and formed tumours in nude mice. An interesting characteristic of the transformed cells was that they became sponta… Show more

“…Cancerrelated activities of MK have been reported by many laboratories. These activities include transforming, migrating, fibrinolytic, mitogenic, antiapoptotic, and angiogenic ones (Kojima et al, 1995;Choudhuri et al, 1997;Kadomatsu et al, 1997;Qi et al, 2000Qi et al, , 2001. Furthermore, we recently succeeded in suppressing tumour growth by using antisense MK oligodeoxyribonucleotide (Takei et al, 2001(Takei et al, , 2002.…”

“…Western blot analysis was performed as described previously. 37 Briefly, we separated the samples by 10% SDS-PAGE and then transferred them to a nitrocellulose membrane (Whatman, Florham Park, NJ). We blocked the membranes with 5% (w/v) dry fat-free milk in PBS with 0.1% Tween for 60 minutes at room temperature.…”

“…(5)(6)(7)(8) Moreover, since both molecules promote cell growth and survival, as well as angiogenesis, it has been suggested that they are involved in carcinogenesis, which is supported by their increased expression in several types of tumors, including osteosarcoma. (9)(10)(11)(12)(13)(14)(15) To analyze the role of Mdk and Ptn in vivo, mouse deficiency models for both genes have been generated. (16)(17)(18) Not necessarily expected, neither the Mdk-nor the Ptn-deficient mice displayed major abnormalities, although a more detailed analysis has demonstrated a role of both molecules in hippocampus development, behavior, and learning.…”
