Traditional olive oil processing includes batch olive paste kneading for 30-60 min. Microwave (MW) treatment of the olive paste have recently shown to continuously provide faster conditioning, while megasound (MS) treatment have been shown to enhance oil extractability. In the present work the virgin olive oil standard parameters, sensory descriptors, and phenolic and headspace composition upon treatment of oils obtained using novel process enhancement interventions, and their combination in an industrial plant is examined. Standard parameters are within the international specifications. The Control and MW process provide similar phenolic composition but a post MS intervention significantly increased total phenolic content, and decrease total C5 and C6 aldehydes in the MW treated paste. These results encourage further developments for a combined continuous MW and megasonic conditioning technology to fasten the olive oil extraction, to enhance yields, and total phenolic content. Practical Applications: Microwave and megasounds are two technologies easily implementable on industrial olive oil extraction plant. They permit to simplify the entire plant and could replace totally the malaxers section. This new technology allows having a uniform paste treatment in continuous mode, enhancing the olive oil quality.

Gynura procumbens (Sambung nyawa) is a bicolor (green and red) herbal plant, which is widely grown in Asia. This herbs are famously used for medicinal purposes as a safe alternative to chemical-based medicine. Recent studies proved that this plant has anti-herpes simplex virus, anti-flammatory and anti-hyperglycaemic properties. This work investigates the extraction of chlorophyll a and b in Gynura procumbens using Solid liquid extraction method. The solid to solvent ratio, temperature, solvent used and extraction time were varied to determine the optimum conditions for extraction. It was found that at 80 C in 90 minutes with 2:5 solid to solvent ratio was the most favorable condition to extract chlorophyll a and b. Five valuable compounds were found from GC-MS analysis, which are 2-hexanal, phenol, oleic acid, copaene and phytol. This implies that Gynura procumbens promises a good source of many useful bioactive compounds.Keywords: Gynura procumbens, sambung nyawa, chlorophyll, extraction, optimization Abstrak Sambung Nyawa ialah pokok herba dwi-warna yang tumbuh secara meluas di Asia. Herba ini sangat popular digunakan sebagai bahan alternatif yang selamat bagi tujuan perubatan berbanding ubatan berunsurkan bahan kimia. Penyelidikan terbaru membuktikan bawaha pokok ini anti-virus herpes simplex, anti-keradangan dan anti-sifat hiperglisemia. Eksperimen ini menyiasat tentang pengekstrakan klorofil a dan b dalam Gynura procumbens. Menggunakan pengekstrakan cecair pepejal. Nisbah cecair pepejal, suhu, pelarut dan masa mengekstrak telah dimanipulasi untuk menentukan keadaan yang optimum untuk proses pengekstrakan tersebut. Keputusan eksperimen mendapati bahawa pengekstrakan selama 90 minit pada suhu 80C serta nisbah cecair pepejal 2:5 merupakan kondisi yang paling sesuai untuk mengekstrak klorofil a dan b. Lima kompoun berharga telah ditemui melalui keputusan analisa GC-MS iaitu 2-heksanal, fenol, asid oleik, copaene and fitol. Keputusan ini mencadangkan potensi Gynura procumbens sebagai sumber yang baik kepada sebatian bioaktif.

The present study aimed to investigate the metabolism of Forsythoside A (FTA) by human fecal bacteria to clarify the relationship between its intestinal metabolism and its pharmacological activities. FTA was incubated with human fecal microflora in vitro to investigate its metabolic process, and highly sensitive and specific ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS) was performed using MetaboLynx software for metabolite analysis. Caffeic acid (CA) and hydroxytyrosol (HT) were obtained by hydrolysis of FTA, and CA was further hydrogenated to form 3,4-dihydroxybenzenepropionic acid (DCA). The anticomplementary, antimicrobial and antiendotoxin activities of FTA and its metabolites by human fecal microflora were evaluated in vitro with a hemolysis assay, the agar disc-diffusion method, the MIC value and the gel clot LAL assay, respectively. The metabolites showed higher biological activity than FTA, especially HT and DCA. Orally administered FTA may be metabolized to HT and DCA, and the pharmacological effects of FTA may be dependent on intestinal bacterial metabolism.
