Microvesicles and Microvesicle-Associated microRNAs Reflect Glioblastoma Regression: Microvesicle-Associated miR-625-5p Has Biomarker Potential

Abstract: Glioblastoma (GB) is the most aggressive and recurrent form of brain cancer in adults. We hypothesized that the identification of biomarkers such as certain microRNAs (miRNAs) and the circulating microvesicles (MVs) that transport them could be key to establishing GB progression, recurrence and therapeutic response. For this purpose, circulating MVs were isolated from the plasma of GB patients (before and after surgery) and of healthy subjects and characterized by flow cytometry. OpenArray profiling and the in… Show more

“…MiR-486-3p displays tumor-suppressive characteristics, being downregulated in hepatocellular carcinoma (HCC) ( Ji et al, 2020 ), retinoblastoma ( Yang et al, 2020 ), osteosarcoma ( Zhang et al, 2021 ), cervical cancer ( Ye et al, 2016 ), adrenocortical carcinoma ( Li et al, 2021a ) ,and oral squamous cell carcinoma ( Garajei et al, 2022 ). However, in glioblastoma ( Simionescu et al, 2022 ), non-small cell lung cancer ( Jin et al, 2022 ), and β-thalassemia ( Lulli et al, 2013 ), it demonstrates an upregulation, thereby fostering disease progression.…”

“…The miRNAs are known as small non-coding RNAs which regulate gene expression post-transcriptionally through the inhibition of translation and/or the degradation of messenger RNA (mRNA) and they have been found in several body fluids, including blood, circulating EVs or associated with lipoproteins or protein complexes [ 17 , 18 ]. The levels of miRNAs are modified during disease progression and miRNAs’ circulating profiles often reflect their modified expression in the tissue of origin or indicate increased intercellular communication [ 19 ]. There are clear data on a possible use of miRNAs as diagnostic biomarkers and therapeutic targets for the endothelial dysfunction in hypertensive patients [ 20 ].…”
