Microtubules and Their Role in Cellular Stress in Cancer

Abstract: Microtubules are highly dynamic structures, which consist of α- and β-tubulin heterodimers, and are involved in cell movement, intracellular trafficking, and mitosis. In the context of cancer, the tubulin family of proteins is recognized as the target of the tubulin-binding chemotherapeutics, which suppress the dynamics of the mitotic spindle to cause mitotic arrest and cell death. Importantly, changes in microtubule stability and the expression of different tubulin isotypes as well as altered post-translation… Show more

“…However, our findings now provide another mechanism through which general Hsp70 inhibitors might be effective anti-cancer agents; by inhibiting Hsc70-mediated regulation of MT stability. Indeed, compounds that alter MT dynamics are an area of extensive study and several are currently used in the clinic (51). Moreover, recent work even implicates tau in the pathogenesis and prognosis of cancer (52)(53)(54)(55)(56), further supporting this mechanistic link between tau, Hsc70 and MTs.…”

“…Coupled with evidence of its aberrant expression levels in a wide range of tumors of neuronal and non-neuronal origin and disease aggressiveness and resistance to chemotherapy, β3-tubulin is thought to be a survival factor. Interaction between this tubulin isotype and VDAC was evoked to explain modulation of metabolic stress and mitochondrial functions in cancer cells (55). Future research will verify the preferential tubulin isotype interaction with VDAC in cells and potential modifications of these interactions under chemotherapeutic treatment.…”

“…More recently, we have also proven that expression of the NKG2D and DNAM-1 activating ligands MICA and PVR, is enhanced by immunomodulatory drugs (IMiDs) ( 24 ). It is well known that microtubules influence a variety of cell stress responses and act as coordinators of cell functions in response to stress ( 25 ). Recent work also suggests that induction of mitotic arrest by inhibitors of microtubule assembly can trigger DNA damage response pathways in mitotic-arrested cells ( [26][27][28] ).…”
