Microtubule cytoskeleton behavior in the initial steps of host cell invasion by Besnoitia besnoiti

Abstract: Besnoitia besnoiti is a protozoan parasite responsible for bovine besnoitiosis. Indirect immunofluorescence showed that isolated B. besnoiti possesses a set of subpellicular microtubules, radiating from the apical end and extending for more than 2/3 of the cell body. Upon interaction with the host cell, B. besnoiti undergoes dramatic modifications of shape and surface, as revealed by atomic force microscopy, accompanied by a distinct tubulin labeling on the posterior region. In the host cell, the microtubule c… Show more

“…Also, for T. gondii-infected host cells, the microtubule cytoskeleton surrounds the parasitophorous vacuole (PV) soon after invasion (Walker et al 2008;Sweeney et al 2010) which is accompanied by the detachment of the centrosome, the major microtubule-organizing centre (MTOC) of animal cells, from the nuclear envelope, and further association with the PV (Coppens et al 2006;Walker et al 2008;Wang et al 2010). For example, Golgi proteins like GM130, Cdc42 and Tuba regulate centrosome morphology and function (Kodani and Sütterlin, 2008;Kodani et al 2009); and a centrosomal protein, like AKAP450, is an important linker between the centrosome and the Golgi apparatus (Rivero et al 2009). However, infected animals were also detected in Portugal (Cortes et al 2006a(Cortes et al , b, 2007Reis et al 2006) without hints, how this disease may have crossed the Mediterranean Sea. Just recently, Fernandez-Garcia et al (2008) succeeded in the first detection of cattle besnoitiosis in Central Spain and described clearly the stages of this parasite during the chronic phase of the disease in a cow and presented tissue cysts, cyst-stages and some of their molecular biological features.…”

“…besnoiti parasites (Bb1Evora03 isolate) used in the preparation of the B-MAT antigen suspension and IFAT slides were obtained from a naturally infected bovine, as described elsewhere (Cortes et al, 2006b;Reis et al, 2006). Parasites were maintained by continuous passage in Vero cell (African Green Monkey kidney epithelial cells) cultures.…”
