Microtubule Alterations and Mutations Induced by Desoxyepothilone B

Abstract: Epothilones, like paclitaxel, bind to beta-tubulin and stabilize microtubules. We selected a series of four leukemia sublines that display increasing levels of resistance to the epothilone analog desoxyepothilone B (dEpoB). The dEpoB cells selected in 30-140 nM were approximately 15-fold cross-resistant to paclitaxel, while 300 nM selected cells were 467-fold resistant to this agent. The dEpoB-selected cells are hypersensitive to microtubule destabilizing agents, and express increased levels of class III beta-… Show more

“…Growth inhibition assays were done as previously described (22). Briefly, cells were seeded at 3,750 per well (HMEC-1 and MRC-5) or 1,500 per well (BMH29L) in 96-well gelatin-coated plates.…”

“…Two-Dimensional PAGE and Immunoblotting Analysis Two-dimensional PAGE of parental CCRF-CEM and 2ME2-resistant cells was carried out as described previously (21), with minor modifications. Isolated microtubule pellets from CCRF-CEM and 2ME2-resistant cells were solubilized with isoelectric focusing buffer (7 mol/L urea, 2 mol/L thiourea, 2% CHAPS, 2% sulfobetaines 3-10, 1% amidosulfobetaine-14, 2 mol/L tributylphosphine, 65 mmol/L DTT, 1% carrier ampholytes 3-10, 1% carrier ampholytes 4-6, 0.01% bromophenol blue).…”

“…5 Furthermore, b-tubulin mutations in these regions have been suggested to be related with tumor cell chemosensitivity to microtubule-binding drugs. 4,6 Paclitaxel is an antineoplastic drug extensively used in breast cancer treatment binding the b-tubulin subunits on the microtubule, blocking cells in G 2 /M phase and leading to apoptotic cell death. 7 These cytotoxic effects have been reported to be clearly related with the induction of microtubule assembly and promotion of microtubule bundle formation.…”
