Microtubule-actin crosslinking factor 1 (Macf1) domain function in Balbiani body dissociation and nuclear positioning

Abstract: Animal-vegetal (AV) polarity of most vertebrate eggs is established during early oogenesis through the formation and disassembly of the Balbiani Body (Bb). The Bb is a structure conserved from insects to humans that appears as a large granule, similar to a mRNP granule composed of mRNA and proteins, that in addition contains mitochondria, ER and Golgi. The components of the Bb, which have amyloid-like properties, include germ cell and axis determinants of the embryo that are anchored to the vegetal cortex upon… Show more

“…Interestingly, we found MACF1 highly enriched in the Buc interactome ( S3 Table ). Zebrafish mutants in macf1 and buc show defects in embryonic polarity and Balbiani body localization [ 67 70 ] supporting the specificity of the biochemical interaction. Another good indicator for the specificity of the pulldown was the detection of the germ plasm component Ziwi (piwil1) [ 34 ], which was not enriched in the Buc sample ( S3 Table ).…”

“…Mice carrying the floxed MACF1 alleles (in B6 background) were acquired through a material transfer agreement between the University of Chicago (USA) and the Northwestern Polytechnical University (China). Construction of the targeting vector was reported previously [5], in which the loxP sites were integrated to excise exons 11,12, and 13 of the Macf1 gene (while in the original literature, the flanked exons were reported as E6 and E7) ( Figure S1A). Mice expressing Cre recombinase under the control of Prx1 (paired related homeobox1) promoter (in B6 background) were acquired from Biocytogen Co., Ltd. (Beijing, China).…”

“…This effect of MACF1 down-expression on mitochondria organization is consistent with a number of previous observations, namely, microtubule network, through molecular motors such as Kif5b and Dynein, previously shown to control mitochondrial movements in myoblasts (iqbal and Hood, 2014). Additionally, MACF1 was shown to affect mitochondria localization through nuclear positioning in oocyte egg (Escobar-Aguirre et al, 2017). Interestingly, the interplay between actin branching and microtubule acetylation has been recently pointed out as a mitochondrial distribution keeper in fibroblasts cells (Shi et al, 2019).…”
