Microtiter Plate-Format Optode

Abstract: Microtiter plate-format optodes could be assembled by casting bulk-response membranes into the standard 96-well polypropylene-based plate or by screen printing them on an optically transparent substrate with 96-well pattern. The compositions of thick optode membranes, especially the ratios of poly(vinyl chloride) (PVC) to plasticizer [bis-(2-ethylhexyl) sebacate (DOS)], were carefully optimized to provide reproducible and rapid response. Adjusting the ratio of PVC to DOS by 1:6, bulk-response membranes contain… Show more

“…In an attempt to increase the throughput of ion determinations, Kim et al have described a 96-well plate-format absorbance-based optode that required microvolume samples and that could be read using existing clinical laboratory instrumentation. 21 An even more promising technique, however, that offers rapid, high-throughput analyses with multiplexing capabilities is flow cytometry. “…To prepare the calcium selective nanosensors, 1.9 mg of NaTFPB, 1.4 mg of Chromoionophore I, 8.0 mg of DOS, 5.0 mg of F127, and 6.5 mg of calcium ionophore IV were dissolved in 3.0 mL of THF to form a homogeneous solution. A 0.5-mL portion of this solution was pipetted and injected into 4.5 mL of deionized water on a vortex with a spinning speed of 1000 r/min.…”

“…In contrast to microelectrodes, they are easy to manufacture, insensitive to stirring, do not consume oxygen and show fairly long-term stability (Klimant et al, 1995). Optodes can be introduced into a variety of closed and flow-through (Sanchez-Pedreno et al, 2000) incubation chambers or even in microtiter plate-format (Kim et al, 1998). They are suited for experimentation under conditions in which conventional chemical analysis or use of polarographic electrodes is difficult, e.g.…”
