Microspectrofluorometry and fluorescence imaging in the study of human cytopathology

Abstract: The study of energy pools and dynamics of specific pathways in living cells by microspectrofluorometry and fluorescence imaging produces spectral and topographic images characterizing structural and functional changes associated with cytopathology. Microspectro‐fluorometry and fluorescence imaging have been applied, together with organelle morphometry to a number of cells mimicking certain cytopathologies, including melanoma cells, long‐term malignant cells, and gene‐defective cells. These investigations of ce… Show more

“…Currently, multispectral imaging is widely applied in biology and pathology (Levenson & Farkas, 1997; Kohen et al, 2000; Levenson, 2004), cytogenetics, histology, and immuno-histochemistry (Liyanage et al, 1996; Schrock et al, 1996; Veldman et al, 1997; Hilgenfeld et al, 2001; Dorritie et al, 2004). In the field of plant biology, the application of quantitative analysis of macromolecules in rice was first reported using the MMI technique by Hu et al (2005).…”

“…Currently, multispectral imaging is widely applied in biology and pathology~Levenson & Farkas, 1997;Kohen et al, 2000;Levenson, 2004!, cytogenetics, histology, andimmuno-histochemistry~Liyanage et al, 1996;Schrock et al, 1996;Veldman et al, 1997;Hilgenfeld et al, 2001;Dorritie et al, 2004!. In the field of plant biology, the application of quantitative analysis of macromolecules in rice was first reported using the MMI technique by Hu et al~2005!. A technique combining the PAS reaction and a microdensitometer was developed to measure the content of the total insoluble polysaccharides during pollen development Pacini & Viegi, 1995;Pardi et al, 1996!. However, a major disadvantage was the inability to obtain information on cellular observations and measurements of insoluble polysaccharide contents simultaneously.…”

“…The studies reported here were carried out by microspectrofluorometry and fluorescence imaging using tricolor and black & white CCD cameras, the latter supplemented by an integrator for image accumulation [1][2]. A Fourier interferometry design is being implemented to measure the excitation and emission spectra simultaneously; the exciting and emission beams are encoded by modulating the light according to wavelength [3].…”
