Microscopic Study of Testicular Tissue Structure and Spermatogenesis Following Long Term Dose Dependent Administration of Monosodium Glutamate in Adult Diabetic Rats

Abstract: Background and aims: Diabetic hyperglycemia leads to structural and functional alterations in body organs including testis. Monosodium glutamate (MSG) is a food

“…Thus, the decrease in testicular weight is related to the decrease in the number of germ and somatic cells in testicles. Comparable results have been reported by Shojaeii et al and Kianifard (Shojaeii et al, 2014;Kianifard, 2016).…”

“…The histopathological lesions in testicular tissues exposed to MSG is similar to those observed by other investigators 49 . The depopulation of germ cells due to inhibition of cell division is responsible for atrophy of ST 87 . The degenerative changes in germ and somatic cells of ST may be arisen from the interaction of MSG with proteins and enzymes interfering with the antioxidant defense mechanism leading to accumulation of free radicals which in turn induces inflammatory reaction and mitochondrial damage 88 .…”

“…Such findings were in consistent with previous studies on the effect of MSG in rat testis. [ 10 36 ] Kianifard[ 37 ] observed reduction in the diameter of STs and decreased number of primary spermatocytes and spermatids following MSG treatment in rats and mentioned that depopulation of germ cells led to tubular atrophy and indicated a reduction of cell divisions. The widening in the intertubular space that was observed in this study might be due to the atrophy and reduced tubular diameter.…”

“…MSG is neurotoxic, when found in high concentration it causes excessive glutamate receptor activation and can induce neuronal death. [ 43 ] Hypothalamic neuronal cell death[ 44 ] and reduction in the level of luteinizing hormone (LH), FSH, and testosterone and decreased sperm count[ 36 37 38 ] were previously reported after administration of MSG in mice and rats. Hence, the damage of hypothalamic neurons by MSG may cause disruption of the hypothalamic–pituitary–gonadal axis that control the normal spermatogenesis and testosterone secretion by the Leydig cells[ 45 ] and can lead to structural and functional alterations of testicular tissue.…”
