Microsatellites from the burbot (Lota lota), a freshwater gadoid fish (Teleostei)

Abstract: We developed 21 polymorphic dinucleotide microsatellite loci, (CA) n and (CT) n , for the Holarctic freshwater fish, Lota lota , using an enriched genomic library protocol. The species has an interesting life history because winter-spawning adults migrate over long distances to form spawning aggregations, a behaviour which should maintain genetic homogeneity across large spatial scales. Availability of the reported microsatellites will facilitate the investigation of population genetic structure with regard to… Show more

“…In agreement with previous studies (Sanetra & Meyer, 2005;Barluenga et al, 2006;Elmer et al, 2008) none Table 1 Hydrobiologia (2012) 683:217-229 221 of the 36 locus pairs shows significant linkage disequilibrium (P [ 0.05), indicating that all loci used in this study can be treated as independent markers. Assignment to subspecies was determined by Structure (see ''Admixture'' below for more details) and these divisions are used for all subsequent analyses.…”

“…All individuals were genotyped at 11 species-specific microsatellite loci (Sanetra & Meyer, 2005;Elmer et al, 2008). As in previous research (Elmer et al, 2008), Llo 32 and Llo 13 were discarded from further analysis because of poor genotype quality.…”

“…Microsatellite markers: A microsatellite-enriched library was prepared from A. burtoni DNA using a magnetic bead enrichment protocol and (CA) 15 and (CT) 15 probes (for a detailed description, see Sanetra and Meyer 2005). A total of 1156 clones were sequenced, and 683 clones (enrichment rate $60%) contained repeat motifs (including $10% duplicate clones).…”

“…Genomic DNA was extracted after a proteinase K digestion using a standard high salt method and ethanol precipitation (Bruford et al 1998). We genotyped all 350 samples at 11 microsatellite loci (Llo1, Llo6, Llo7, Llo12, Llo13, Llo15, Llo16, Llo21, Llo26, Llo32, Llo34) following established methods (Sanetra and Meyer 2005). Loci were amplified in three separate multiplex reactions using a multiplex polymerase chain reaction (PCR) kit (Qiagen) following manufacturer's protocols and using 0.1-0.2 mmolÁL -1 of each primer per reaction.…”
