MicroRNAs interfere with DNA methylation in rheumatoid arthritis synovial fibroblasts

Abstract: BackgroundThe DNA of rheumatoid arthritis synovial fibroblasts (RASF) is globally hypomethylated; this contributes to an aggressive behaviour. In an attempt to remethylate these cells, we supplemented with methyl donors. We investigated the possible interference of microRNAs (miRs).Material and methodsRASF were treated with L-methionine or betaine. Transcripts of de novo methyltransferases (DNMTs) and miRs were measured by real-time PCR, and a transcription PCR array was performed. Levels of homocysteine, matr… Show more

“…In addition to ER stress, betaine also inhibits apoptosis. In a recent study of rheumatoid arthritis synovial fibroblasts, Gaur and colleagues found that transcription factor-3 (ATF-3), an apoptosis-related molecule, is downregulated by betaine ( 131 ). In addition, betaine can inhibit caspase family proteins.…”

“…MiRNAs post-transcriptionally regulate the degradation and translation of their target genes. MiRNAs are involved in the inflammatory response and the development of autoimmune diseases, such as rheumatoid arthritis [14], and multiple sclerosis [15]. Increasing evidence has shown that aberrant miRNA expression is linked to the regulation of autophagy [16].…”

“…The DNA methyltransferase (DNMT)3A was defined as a target of miR-29 in RA-FLS. Thus, miR-29 might repress the remethylation effect of bethaine [40]. Miao et al showed that miR-152 was downregulated in an arthritis rat model, and the overexpression of miR-152 in FLS decreased the expression of DNMT1.…”
