MicroRNAs 10a and 10b Regulate the Expression of Human Platelet Glycoprotein Ibα for Normal Megakaryopoiesis

Abstract: MicroRNAs are a class of small non-coding RNAs that bind to the three prime untranslated region (3′-UTR) of target mRNAs. They cause a cleavage or an inhibition of the translation of target mRNAs, thus regulating gene expression. Here, we employed three prediction tools to search for potential miRNA target sites in the 3′-UTR of the human platelet glycoprotein (GP) 1BA gene. A luciferase reporter assay shows that miR-10a and -10b sites are functional. When miR-10a or -10b mimics were transfected into the GP Ib… Show more

“…In addition, the levels of miRNAs that had been previously shown to regulate platelet membrane proteins were measured. These include mir-10a, which regulates the expression of glycoprotein 1b (GP1b) [21], and mir-223, which controls the expression of putative G protein coupled receptor (P2Y12) [22, 23]. We also measured the level of mir-376c, which modifies the protease activated-receptor 4 (PAR4) signaling via phosphatidylcholine transfer protein (PCTP) [24].…”

“…Importantly, platelet miRNAs are known to regulate platelet protein expressions [21, 22, 24]. For instance, a sequential increase in mir-326 expression during the storage period down-regulates the expression of the anti-apoptotic gene BCL-XL and results in platelet apoptosis [29].…”

“…These miRNAs have been identified with inactivation and other associated functions such as reactivity, hemostasis, differentiation and apoptosis. For example, platelet activation-related miRNAs have been identified with differential expression analysis including miR-10a, miR-10b, miR-24, miR-145, miR-183, miR-197, miR-21, miR-26a-1, miR-27b, miR-411, miR-423, and miR-451a [19,24,27,34,35,36,39,40].…”

“…A limitation of this study is the relatively small number of miRNAs investigated, only 20 more expressed, among which, some have been intensively reviewed by the literature [ 7 , 30 , 64 , 65 , 66 ]. Our results also provided a panel of miRNAs that have had their functions elucidated in platelets such as miR-223 [ 24 , 66 ], miR-20a [ 23 ] miR-126 [ 67 ], miR-10a/miR-10b [ 68 ], miR-326 [ 69 ], miR-150/miR-501/miR-338-5p/miR-432-5p/miR-411-5p [ 21 ], miR-570 [ 18 ], miR-495 [ 70 ], and miR-21/miR-27b [ 71 ]. For example, specific cold storage conditions caused changes in miRNA expressions, increasing levels of miR-20a/miR-10a/miR-16-2/miR-223, which correlated with the quality of platelets under specific conditions of storage [ 23 ].…”
