Scientific evidence supports the early deregulation of epigenetic profiles during breast carcinogenesis. Research shows that cellular transformation, carcinogenesis, and stemness maintenance are regulated by epigenetic-specific changes that involve microRNAs (miRNAs). Dietary bioactive compounds such as blueberry polyphenols may modulate susceptibility to breast cancer by the modulation of CSC survival and self-renewal pathways through the epigenetic mechanism, including the regulation of miRNA expression. Therefore, the current study aimed to assay the effect of polyphenol enriched blueberry preparation (PEBP) or non-fermented blueberry juice (NBJ) on the modulation of miRNA signature and the target proteins associated with different clinical-pathological characteristics of breast cancer such as stemness, invasion, and chemoresistance using breast cancer cell lines. To this end, 4T1 and MB-MDM-231 cell lines were exposed to NBJ or PEBP for 24 h. miRNA profiling was performed in breast cancer cell cultures, and RT-qPCR was undertaken to assay the expression of target miRNA. The expression of target proteins was examined by Western blotting. Profiling of miRNA revealed that several miRNAs associated with different clinical-pathological characteristics were differentially expressed in cells treated with PEBP. The validation study showed significant downregulation of oncogenic miR-210 expression in both 4T1 and MDA-MB-231 cells exposed to PEBP. In addition, expression of tumor suppressor miR-145 was significantly increased in both cell lines treated with PEBP. Western blot analysis showed a significant increase in the relative expression of FOXO1 in 4T1 and MDA-MB-231 cells exposed to PEBP and in MDA-MB-231 cells exposed to NBJ. Furthermore, a significant decrease was observed in the relative expression of N-RAS in 4T1 and MDA-MB-231 cells exposed to PEBP and in MDA-MB-231 cells exposed to NBJ. Our data indicate a potential chemoprevention role of PEBP through the modulation of miRNA expression, particularly miR-210 and miR-145, and protection against breast cancer development and progression. Thus, PEBP may represent a source for novel chemopreventative agents against breast cancer.

The Standardized Cultured Extract of Lentinula edodes Mycelia (also known as Active Hexose Correlated Compound, AHCC) and Wasabia japonica (Wasabi) are natural nutritional supplements known for their immunomodulatory and anticancer potential. The aim of this study was to evaluate the combinatorial effect of the bioactive immunomodulatory compound (BAIC), obtained by combining Wasabi and AHCC, on human breast (MCF-7) and pancreatic (Panc02) adenocarcinoma cell lines. Data obtained revealed that BAIC determines a striking decline in cancer cell growth at minimal concentrations compared with the use of Wasabi and AHCC as single agents. A significant increase in the G0/G1 subpopulation together with a marked augmentation in the percentage of apoptotic cells was demonstrated by flow cytometry, together with a significant upregulation in the expression of genes associated to the apoptotic cascade in both cell lines. The inhibitory role BAIC plays in mammospheres formation from MCF-7-derived cancer stem cells was shown with a marked reduction in size and number. Interestingly, when BAIC was exposed to monocytic cells, no cytotoxic effects were observed. A monocytes-to-macrophages differentiation was rather observed with the concomitant acquisition of an anti-inflammatory phenotype. Taken together, our findings suggest that BAIC could be used as a potential integration of standard chemotherapy treatments because of the improved inhibitory activity on cancer cell proliferation and reduced potential adverse effects.

Background: A standardized extract of cultured Lentinula edodes mycelia (ECLM), an extract from cultured Lentinula edodes, has been reported to suppress breast cancer stem cell proliferation by regulating microRNA (miR) expression. Natural antisense RNAs (ASs), a type of protein non-coding RNA, can regulate the expression of protein-coding genes by acting as a competing endogenous RNA (ceRNA) that adsorbs miRNAs, resulting in the prevention of mRNA degradation, and can also form a transient RNA duplex with mRNA. EphA2, a receptor tyrosine kinase, is typically expressed at low levels in normal epithelial cells, whereas its overexpression has been widely observed in numerous solid tumors and is associated with cell transformation, primary tumor initiation, and tumor progression. Objective: This study aimed to investigate the effect of ECLM on the expression of both EphA2 mRNA and endogenous AS to this mRNA, which could negatively affect human breast carcinoma cell proliferation. Methods: We used MCF7 and MDA-MB-231 human breast carcinoma cells, which were sub-cultured three times in the presence of optimized concentrations of ECLM. The effect of ECLM on the expression of EphA2 AS and mRNA was analyzed by RT-qPCR. miRNAs targeting both EphA2 AS and EphA2 mRNA and their RNA miR response elements (MREs) were predicted and analyzed by RT-qPCR and luciferase reporter assays. Results: ECLM suppressed the proliferation of MCF7 and MDA-MB-231 cells in a dose-dependent manner. In cells for which proliferation was negatively affected by ECLM, EphA2 AS and mRNA expression was also significantly inhibited by ECLM. Although neutralization of miR-335 led to the de-repression of both EphA2 AS and mRNA, results did not fully support the possibility that EphA2 AS might function as a ceRNA to regulate EphA2 mRNA levels. Conclusion: ECLM suppressed the proliferation of breast carcinoma cells in a specific dose-dependent manner. This suppressive effect was associated with a concordant reduction in both EphA2 AS and mRNA expression. These effects were not thought to occur via the reported ceRNA effect. These results thus suggest that ECLM could regulate EphA2 AS and mRNA expression by forming a transient RNA duplex formation, thereby stabilizing EphA2 mRNA.
