MicroRNA (miR) 125b regulates cell growth and invasion in pediatric low grade glioma

Abstract: Members of the miR-125 family are strongly expressed in several tissues, particularly brain, but may be dysregulated in cancer including adult and pediatric glioma. In this study, miR-125 members were downregulated in pilocytic astrocytoma (PA) as a group compared to non-neoplastic brain in the Agilent platform. In the Nanostring platform, miR-125 members were downregulated primarily in pleomorphic xanthoastrocytomas and gangliogliomas. Using CISH for miR-125b, highest levels of expression were present in grad… Show more

“…Furthermore, we also demonstrated that miRNAs hsa-miR-199a-3p and hsa-miR-140-3p negatively regulate COX-2 and ADAMTS-5 expression, respectively in human OA chondrocytes 14,15 . Recently, the function of miR-125b was discovered in various cell types and its association with several human disorders was reported 1619 . In OA, the function of miR-125b was somewhat defined by few studies only, in one study miR-125b was reported to regulate aggrecanase-1 expression in human chondrocytes 20 and in another study, miR-125b was reported to regulate the inflammatory activities in stimulated chondrogenic cells via MIP-1α signaling event 21 .…”

“…Also, miR-125 was upregulated in esophageal adenocarcinoma and associated with poor prognosis [24]. However, miR-125 was under-expressed in glioma and regulated cell growth and invasion [25]. Besides, Chen H showed that miR-125 downregulation acted as a potential biomarker for colorectal cancer treatment [26].…”

“…A single miRNA can regulate different targets, and it is estimated that miRNAs regulate approximately 60% of all human protein coding genes by post-transcriptional mechanisms [ 22 ]. Therefore, miRNAs are involved in several biological processes, including apoptosis [ 23 ], cell metabolism, homeostasis [ 24 ], differentiation [ 25 ], cell cycle [ 26 ], growth [ 27 ], cell survival [ 28 ], cell proliferation, and migration [ 29 ]. In addition, the involvement of miRNAs has also been demonstrated in pathological processes of different diseases, such as heart failure [ 30 ], arterial hypertension [ 31 ], type 2 diabetes mellitus, obesity [ 32 ], Chagas disease cardiomyopathy [ 33 ], and AD [ 34 ].…”
