MicroRNA-mediated regulation of T helper cell differentiation and plasticity

Abstract: T helper (TH) cells orchestrate appropriate cellular and humoral immune responses to a wide variety of pathogens and are central to the success of vaccines. However, their dysregulation can cause allergies and autoimmune diseases. The TH cell universe is characterized by a diversity of distinct cell types, such as TH1, TH2, TH17 cells, regulatory T cells, and T follicular helper cells, each featuring specific functions and gene expression programs, but also by plasticity among the different TH cell subsets. He… Show more

“…Although not covered here, it is important to consider that T cell plasticity is regulated by micro-RNA-mediated post-transcriptional regulation of a wide variety of genes -such as those involved in cytokine signalling, TCR and co-stimulatory signalling, cytosolic signalling and transcriptional regulatory pathways 163 . 169 and IL-17-expressing T H 2 cells in atopic asthma 13 .…”

“…Recently, numerous studies regarding the role of miRNAs in development and function of immune cells have emerged, and several excellent reviews suggest that miRNAs represent a novel layer of control over cellular physiology [1][2][3]. T cells that express the CD8 coreceptor and recognize peptide-MHC class I complexes have a key role in the control of numerous viral or intracellular bacterial infections [4].…”

“…Helper T cells play an essential role in regulating the immune response, and a great deal of effort has been undertaken to understand their development and function (illustrated in several recent reviews [1][2][3] ). However, many questions remain, and many approaches have been utilized to study the mechanisms controlling helper T cell development and function.…”

“…In addition miRNAs are important later on in the development of many tissues. They are thought to function by fine-tuning the expression of genes required for developmental programs 1 . In helper T cells miRNAs play multiple roles and are required for regulatory T cell (Treg) development [11][12][13][14] .…”
