MicroRNA Expression Profiles in Superficial Esophageal Squamous Cell Carcinoma before Endoscopic Submucosal Dissection: A Pilot Study

Abstract: Esophageal squamous cell carcinoma (ESCC) has a poor prognosis when diagnosed at an advanced stage, and early detection and treatment are essential to improve survival. However, intraobserver and interobserver variation make the diagnosis of superficial ESCC difficult, and suitable biomarkers are urgently needed. Here, we compared the microRNA (miRNA) expression profiles of superficial ESCC tissues and adjacent normal tissues obtained immediately before esophageal endoscopic submucosal dissection. We found tha… Show more

“…The most recent findings on the potential benefits of mRNAs indicate they may serve as prognostic factors in ESCC [49] . For instance, miR-10, miR-18, miR-19, miR-21, miR-25, miR-31, miR-129, miR-365, miR-451, miR-1246, and miR-1322 are upregulated in tumor tissues of ESCC patients compared with normal tissues; moreover, miR-155, miR-203, miR-205, miR375, and miR-718 are downregulated in ESCC patients compared to control groups 50 , 51 . Furthermore, several investigations have determined that miRNAs could be accessible noninvasive biomarkers to monitor responses to therapy, such as radiotherapy and chemotherapy.…”

“…Biopsy samples of the tumor and adjacent normal tissue were taken from each patient immediately before completing ESD. The sampling procedure was the same as that described in our previous study (12). All tissue samples were frozen immediately after excision and in vivo 37: 1980-1990 (2023) 1981 1,525760 0,000372 0,010005 Down-regulated hsa-miR-133b 0,324593 0,000013 0,001033 hsa-miR-133a-3p 0,380456 0,000151 0,004900 hsa-miR-378g 0,444162 0,000005 0,001033 hsa-miR-378c 0,452104 0,000016 0,001033 hsa-miR-378d 0,454092 0,000019 0,001033 hsa-miR-378e 0,464034 0,000015 0,001033 hsa-miR-378f 0,467831 0,000009 0,001033 hsa-miR-378b 0,469599 0,000001 0,000433 hsa-miR-378i 0,470580 0,000012 0,001033 hsa-miR-572 0,479847 0,000412 0,010528 hsa-miR-378a-5p 0,516377 0,000044 0,001854 hsa-miR-3185 0,555634 0,000279 0,008322 hsa-miR-378a-3p 0,558695 0,000002 0,000677 hsa-miR-638 0,610290 0,000324 0,009267 hsa-miR-4731-5p 0,647158 0,000019 0,001033 Fold change (FC)>1.5 and FC<0.67, p<0.001.…”

“…MiRNAs can be obtained not only from body fluids but also from tissues. Fujihara et al [ 58 ] obtained matched sets of primary ESCC tissue samples and corresponding neighboring healthy tissues from patients with a pathological diagnosis of stage I ESCC. Then, unsupervised hierarchical cluster analysis was used to determine the variation in gene expression patterns between ESCC tissues and neighboring healthy tissues, and 48 differentially expressed miRNAs were revealed.…”
