MicroRNA Expression in Melanocytic Nevi: The Usefulness of Formalin-Fixed, Paraffin-Embedded Material for miRNA Microarray Profiling

Abstract: MicroRNAs (miRNAs) are small, noncoding RNA molecules that regulate cellular differentiation, proliferation, and apoptosis. MiRNAs are expressed in a developmentally regulated and tissue-specific manner. Aberrant expression may contribute to pathological processes such as cancer, and miRNA may therefore serve as biomarkers that may be useful in a clinical environment for diagnosis of various diseases. Most miRNA profiling studies have used fresh tissue samples. However, in some types of cancer, including malig… Show more

“…Storage up to 7 years did not cause significant deterioration of miRNA quality, but older samples may exhibit loss of small RNA quality due to oxidation and fixation in nonbuffered formalin. 24 Formalin-fixed paraffin-embedded samples have already been successfully used for miRNA expression profiling in various tissues, 15,25,26 but there have been no data on their applicability for miRNA expression analysis in pheochromocytomas. We have observed good correlation between miRNA microarray results retrieved from three matched frozen and formalin-fixed paraffin-embedded samples derived from the same pheochromocytoma tissues.…”

“…2 Many subsequent reports of miRNAs in cancer have been reviewed. 3 In melanoma, miRNAs can be readily analyzed from formalin-fixed paraffin-embedded tissue, 4 show high frequency genetic alterations, 5 and exhibit a melanoma-specific signature. 6 In addition, miRNAs have shown prognostic value in melanoma.…”
