MicroRNA-directed pathway discovery elucidates an miR-221/222–mediated regulatory circuit in class switch recombination

Abstract: MicroRNAs (miRNAs, miRs) regulate cell fate decisions by post-transcriptionally tuning networks of mRNA targets. We used miRNA-directed pathway discovery to reveal a regulatory circuit that influences Ig class switch recombination (CSR). We developed a system to deplete mature, activated B cells of miRNAs, and performed a rescue screen that identified the miR-221/222 family as a positive regulator of CSR. Endogenous miR-221/222 regulated B cell CSR to IgE and IgG1 in vitro, and miR-221/222–deficient mice exhib… Show more

“…A recent study proved evidence that it’s also critical in JIA-associated autoimmune uveitis ( 53 ). It is worth noting that ARID1A was also found playing important role in T-cell development ( 54 , 55 ), while its expression in B cells was found to regulate levels of IgE and IgG ( 56 ). Moreover, ARID1A has been found to induce antiviral IFN-I production in macrophages as well ( 57 ).…”

“…We found that the risk allele for rheumatoid arthritis was correlated with high expression levels of AFF3 and frequent utilization of IgG2 and IgA2 in B cells. In patients with rheumatoid arthritis, rheumatoid factor IgG and anticyclic citrullinated peptide IgG antibodies are detected even before the onset of the disease and are related to disease severity, suggesting that specific classes of autoantibodies are related to the pathogenesis (60)(61)(62)(63). Moreover, islet-specific autoantibodies (anti-insulin, glutamate decarboxylase, islet antigen 2, and islet-specific zinc transporter antibodies) have been detected in patients with T1D (64)(65)(66) and are considered to be the best available markers for predicting the disease.…”

“…As in all cells, microRNAs are important determinants of B-cell function and patterned responses [83]. As many miRNAs are ROS-regulated, alterations in mitochondrial function and mitochondrial ROS production will drive variations in miRNAs, thereby shaping patterned gene expressions in B-cells.…”
