MicroRNA detection on microsensor arrays by SPR imaging measurements with enzymatic signal enhancement

Abstract: We investigated sequence-specific and simultaneous microRNA (miRNA) detections by surface plasmon resonance (SPR) imaging measurements on SPR chips possessing an Au spot array modified with probe DNAs based on a miRNA-detection-selective SPR signal amplification method. MiRNAs were detected with the detection limit of the attomole level by SPR imaging measurements for different miRNA concentrations on a single chip. SPR signals were enhanced based on a combination process of sequence-specific hybridization of … Show more

“…Most miRNA genes exist in the genome as a single copy, multiple copies, or in a cluster [7]. miRNAs play important roles in various cellular functions, such as proliferation, invasion, and migration [8]. Furthermore, they have been reported to be closely associated with many kinds of tumors including glioma [9].…”

“…(10,25,26) However, simple, rapid, and "one-time" disposable devices to avoid false positive results without any labeling of targets or any external indicators are required for reliable oligonucleotide detection in the environmental and biomedical fields. (17,27) Our technique might contribute to developing practical oligonucleotide sensor array chips that can make environmental and biomedical diagnoses simpler and easier to perform.…”

“…Attributed to the sensitive signal transducing, SPR and SERS techniques have been adapted towards EV RNA evaluation. In fact, SPR based RNA biosensors have been developed with a variety of sophisticated plasmonic probes and signal transduction techniques (Aoki et al., 2019; Coutinho & Somoza, 2019; Fong & Yung, 2013; Xue et al., 2019), and such methods were applied for analyzing assorted biological samples, for instance, urinary miRNA (Yeung et al., 2018) and Zika viral RNA (Adegoke et al., 2017). However, few published investigations have focused on EV RNA characterization.…”
