MicroRNA: Crucial modulator in purinergic signalling involved diseases

Abstract: Both microRNAs (miRNAs) and purinergic signalling are widely and respectively expressed in various tissues of different organisms and play vital roles in a variety of physiological and pathological processes. Here, we reviewed the current publications contributed to the relationship of miRNAs and purinergic signalling in cardiovascular diseases, gastrointestinal diseases, neurological diseases, and ophthalmic diseases. We tried to decode the miRNAs-purinergic signalling network of purinergic signalling involve… Show more

“…MicroRNAs (miRNAs) are endogenous 22-24nt non-coding single-stranded RNA molecules that regulate post-transcriptional gene expression (He and Hannon, 2004). Both miRNAs and purinergic signaling are widely expressed in various tissues of different organisms, and they play important roles in various physiological and pathological processes (Guo et al, 2023). Zhou L et al proved that increased expression of miRNA-186 and miRNA-150 in cancer epithelial cells reduces P2X7R mRNA by binding to the 3' untranslated region (3'-UTR) of P2X7R, regulating cell growth and apoptosis (Zhou et al, 2008).…”

“…P2RX7 has been shown to be crucial for the regulation of various signaling pathways, such as the inflammasome pathway or those that lead to the release of cytokines, cell death or mitochondrial activation (3)(4)(5)(6). Next to its contribution to various pathological diseases, P2RX7 activation is also associated with the release of extracellular vesicles (EVs) or particles (EPs) (7)(8)(9). They contain various immunostimulatory molecules known to be pivotal for the activation of antigen presentation processes (10,11).…”

“…Moreover, nanoparticles can assist with improved delivery of drugs such as A 2A antagonists that counteracted immunosuppression ( Arruga et al, 2021 ). It is notable that the purinergic signaling network is subjected to modulation by microRNA (miRNA) ( Ferrari et al, 2016 ), and over 30 miRNAs directly or indirectly modulate P1 and P2 receptors and ectoenzymes, with miR-187 capable of modulating both P2X7 and CD73 ( Guo et al, 2022 ). Notably, miRNA that bind the 3′ untranslated region of the P2X7 receptor can affect the development of breast cancer by influencing the P2X7 receptor expression ( Zhu et al, 2021 ).…”
