MicroRNA-9 and MicroRNA-326 Regulate Human Dopamine D2 Receptor Expression, and the MicroRNA-mediated Expression Regulation Is Altered by a Genetic Variant

Abstract: Background: Regulation of dopamine D2 receptor (DRD2) is pathophysiologically and pharmacologically important. Results: miR-9 and miR-326 target to the 3Ј-UTR of DRD2, and endogenously inhibit DRD2 expression. A functional single nucleotide polymorphism alters such regulation. Conclusion: DRD2 is post-transcriptionally regulated by miR-326 and miR-9. Significance: The study suggests a pathophysiological and pharmacological role of miR-9 and miR-326 in neuropsychiatric disorders.

“…The dopamine receptor D2 (DRD2) gene, a downstream target of miR-9, is also implicated in the pathophysiology of schizophrenia. The SNP rs1801028 (Cys311Ser) in DRD2 is linked to schizophrenia in a metaanalysis (Glatt et al, 2003;Shi et al, 2014). An inverse correlation in the expression of DRD2 and miR-9 has been observed in regions of human brains implicated in schizophrenia (Shi et al, 2014).…”

“…Gene expression is regulated at the transcriptional and post-transcriptional (RNA decay and protein synthesis) levels, so noncoding variants can influence gene expression through both transcriptional and post-transcriptional regulatory mechanisms [3032] . Compared to RNA decay, transcription remains the predominant mechanism determining individual expression variation [31] .…”

“…TaqMan genotyping of chr20_43249673_CTCTT_del in the MGS EA sample (2663 cases and 2653 controls) was performed as described previously (72,73) on LightCycler 480 II (Roche). We did not genotype chr20_43254291_T_ins, because the TaqMan custom assay (Life Technologies) could only be designed for chr20_43249673_CTCTT_del.…”
