MicroRNA‐7 negatively regulates Toll‐like receptor 4 signaling pathway through FAM177A

Abstract: Toll-like receptor (TLR) 4 signalling is critical for innate immunoinflammatory response and widely triggers the development of various types of clinical diseases. MicroRNA-7 (miR-7) is well documented to play an important regulatory role in various biological events. However, the exact role of miR-7 in TLR4 signalling pathway remains to be fully elucidated. In the present study, we found that miR-7 expression in TLR4 signallingactivated bone marrow-derived macrophages (BMDMs) stimulated by LPS was dramaticall… Show more

“…In Tyrobp –/– mice, Pcdhgb4 and Pcdhga8 were upregulated, while Pcdhga2, Pcdhga9 and Pcdhgb8 were downregulated, suggesting that Pcdhg gene expression might have some relation with TYROBP. Fam177a was upregulated in our Tyrobp –/– mice, and downregulation of this gene impairs the transduction of TLR4 signaling; inhibits the phosphorylation of NF-κB, Akt, Erk1/2 and JNK; and downregulates TLR4, MyD88, TRAF6, and TRIF ( Chen et al, 2021 ). The potential implication of Fam177a in AD requires further investigation.…”

“…Inflammation is one of the markers of accelerated aging and age-related diseases [ 135 , 136 , 137 ]. Meaningfully, the let-7 family also plays a key role in regulating immune-mediated inflammation [ 138 ].…”

“…miR-99a promotes the differentiation of Treg cells [ 52 ], and miR-199 is involved in the development of Th17 and Treg cells in patients with multiple sclerosis [ 53 ], showing increased expression in some autoimmune diseases, such as atopic eczema and SLE [ 54 ]. Moreover, the expression of miR-7 was found to be increased during the activation of TLR4 signaling, suggesting that it could act as a new negative fine-tuner in the regulation of its signaling [ 55 ].…”
