MicroRNA-451 inhibits inflammation and proliferation of glomerular mesangial cells through down-regulating PSMD11 and NF-κB p65

Abstract: The present study aimed to investigate the regulatory roles of microRNA-451 (miR-451) on the inflammation and proliferation of glomerular mesangial cells (GMCs) under high-glucose condition, and reveal the potential mechanisms related to 26S proteasome non-ATPase regulatory subunit 11 (PSMD11) and nuclear factor-κ B (NF-κB) signaling. The interaction between PSMD11 and miR-451 was identified by dual luciferase reporter (DLR) gene assay. GMCs were treated with 5.6 mmol/l (normal, L-GMCs) and 30 mmol/l glucose (… Show more

“…Besides, miR-451 expression was found to increase in the exosomes secreted by the hPT cells in response to injury; however, high glucose treatment for 48 h failed to elicit this response. In response to high glucose, marked downregulation of miR-451 has been shown in mesangial cells (Sun et al, 2016;Wei et al, 2019). We also found that longer exposure of proximal tubule cells to high glucose conditions (for 72 h) led to a rise in miR451 in these cells and their derived exosomes.…”

“…2). miR-451 transfection in glomerular mesangial cells under high-glucose conditions was shown to decrease IL-1b, IL-6, IL-8, and COX-2 46 . The differences in cell type and glucose concentration may account for these differences.…”

“…In recent years, several studies have reported therapeutic strategies targeting Rpn6. Heitmeier and co-workers reported the increase in the assembly of 26S proteasomes accompanied by an elevated expression of Rpn6 in right ventricular hypertrophy (RVH) of pulmonary hypertension (PH), indicating that Rpn6 is a novel potential target to specifically interfere with 26S proteasome activation in RVH . Recently, Wu et al studied the regulatory mechanism of Rpn6 in age-related hearing loss (ARHL).…”
