Lung cancer is the leading cause of cancer-related deaths, of which most can be attributed to non-small cell lung cancer (NSCLC). microRNAs (miRNAs) are a group of small non-coding RNAs that focus on post-transcriptional modification. The present study aimed to investigate the role and function of microRNA-370 (miR-370) in NSCLC and explore the underlying functional mechanisms. We found that miR-370 was significantly downregulated in the tumor tissues of NSCLC patients as well as in NSCLC cell lines. Overexpression of miR-370 by infection of recombinant lentivirus markedly inhibited cell proliferation and promoted cell apoptosis of NSCLC cells. In addition, in vivo tumor formation of NSCLC cells was decreased by miR-370 overexpression. Through bioinformatic analysis, we found that tumor necrosis factor receptor-associated factor 4 (TRAF4), an oncogene as previously reported, was predicted as a putative target gene of miR-370. The direct targeting relationship between miR-370 and the 3'-untranslated region was validated by dual-luciferase reporter assay. Furthermore, overexpression of miR-370 downregulated the protein expression of TRAF4 in the NSCLC cells. Moreover, the growth inhibitory effect of miR-370 overexpression on NSCLC cells was abrogated by TRAF4 overexpression. In conclusion, our results suggest that miR-370 plays an important role in NSCLC by regulating TRAF4 and may be a potential target for the treatment of NSCLC.

Background: Lung cancer is the most common cancer worldwide. The most frequent type of lung cancer is non-small cell lung cancer (NSCLC). MicroRNAs (miRNAs) have been reported to play important role in human cancers. Studies suggest that the aberrant expression of miRNAs could act as the diagnostic or prognostic biomarker in human cancers, including lung cancer. MicroRNA-302b (miR-302b) has ever been investigated in several human cancers. The aim of this study was to examine the prognostic value of miR-302b in patients with NSCLC.Methods: Quantitative real-time RT-PCR (qRT-PCR) analysis were used to measure the expression level of miR-302b in NSCLC and adjacent noncancerous samples. The relationship of miR-302b with the clinicopathological data of NSCLC was analyzed by Chi-square test. The prognostic value of miR-302b was assessed by using the Kaplan-Meier survival curves and Cox regression analysis.Results: The expression level of miR-302b was downregulated in the NSCLC samples compared with the paired adjacent noncancerous samples (P < 0.05). The decreased miR-302b was found correlated with the differentiation (P = 0.019) and lymph node metastasis (P = 0.019) of NSCLC. The survival curves suggested that patients with lower miR-302b expression had poor overall survival than those with high miR-302b expression. The results of Cox analysis demonstrated that the expression of miR-302b was an independent and effective prognostic factor in NSCLC patients with the P of 0.002 (HR = 2.508, 95% CI = 1.410 - 4.463).Conclusion: In one word, the expression of miR-302b was decreased in NSCLC samples, and the miR-302 expression might act as a prognostic biomarker in NSCLC patients.

Background: Lung cancer is one of the most common causes of cancer death among all the malignancies worldwide. Evidences suggest that the incidence and mortality of lung cancer has been on the rise. MicroRNA-449a (miR-449a) as one important member of microRNAs, has been demonstrated acting as a tumor suppressor in lung cancer. In this study, we sought to assess the relationship between miR-449a expression level and diagnostic value of lung cancer.Methods: In this present research, quantitative Real-Time PCR was applied to detect the miR-449a expression in 116 lung cancer patients and 41 healthy volunteers. The diagnostic value of miR-449a in lung cancer patients was determined by receiver operating characteristic (ROC) curve.Results: MiR-449a was significantly down-regulated in lung cancer patients compared with healthy control (P<0.05). In addition, miR-449a expression was associated with sex (P=0.004), tumor size (P=0.000), TNM stage (P=0.006) and metastasis (P=0.036). However, there was no correlation with age, smoking history and histological type of lung cancer patients (all P＞0.05). In the ROC analysis, the results showed that the area under the ROC curve (AUC) was 0.902 with the sensitivity of 94.8% and specificity of 78.0%, and the optimum cutoff value was 2.255.Conclusion: MiR-449a expression was down-regulated in lung cancer patients, and it could be an efficient diagnostic biomarker in lung cancer patients.
