MicroRNA-375 Is Downregulated in Gastric Carcinomas and Regulates Cell Survival by Targeting PDK1 and 14-3-3ζ

Abstract: We investigated expression profiles of microRNA (miRNA) in gastric carcinomas by use of a miRNA microarray platform covering a total of 470 human miRNAs. We identified 39 differentially expressed miRNAs in gastric carcinoma, of which six were significantly downregulated and the other 33 were upregulated. We found that miRNA-375 (miR-375) was the most downregulated and that its ectopic expression in gastric carcinoma cells markedly reduced cell viability via the caspase-mediated apoptosis pathway. Interestingly… Show more

“…The hsa-mir-638 is one of the miRNAs most abundantly expressed in normal serum. Although the physiologic significance of has-mir-638 is not adequately known, one report shows decreased expression of hsa-mir-638 in gastric cancer tissues (44). Thus, our data suggest that hsa-mir-638 may be a candidate for a new therapeutic target in gastric cancer.…”

“…Although several studies did not notice an upregulation of miR-375 in prostate carcinoma samples, various studies observed increased levels of this miRNA in prostate cancer serum samples (44,45). Publications confirm that miR-375 is downregulated in other cancer types such as cervical, lung, or head and neck carcinoma, as well as glioma (46)(47)(48)(49)(50) but upregulated in breast carcinoma (51). Likewise, miR-15a was previously shown to be downregulated in metastasizing prostate carcinoma (18).…”

“…Recently, miR-375 hypermethylation was reported in breast cancer and gastric cancer [35,36]. In breast cancer, higher expression of miR-375 in ERa-positive breast cell lines was shown to influence cell proliferation, and miR-375 overexpression caused loss of epigenetic marks including H3K9me2 and local DNA hypomethylation.…”

“…In gastric cancer, miRNA-375 was found to be the most down-regulated miRNA, and ectopic expression of miRNA-375 in gastric carcinoma cells markedly reduced cell viability by a mechanism involving the caspase-mediated apoptosis pathway [37]. Interestingly, the authors reported that expression of miR-375 inhibited expression of PDK1, a direct target of miR-375, and resulted in the suppression of Akt phosphorylation [36].…”
