MicroRNA-30d Induces Insulin Transcription Factor MafA and Insulin Production by Targeting Mitogen-activated Protein 4 Kinase 4 (MAP4K4) in Pancreatic β-Cells

Abstract: Background: miR-30d induces insulin production, but the underlying mechanism remains unexplored. Results: miR-30d activates MafA by targeting TNF-␣-activated MAP4K4. Conclusion: miR-30d promotes insulin production and protecting ␤-cell functions impaired by proinflammatory cytokines. Significance: Overexpression of miR-30d would be beneficial in preventing the development of diabetes.

“…miRNAs are small (approximately 22 nucleotides in length), noncoding singlestranded RNA molecules that can regulate numerous biological processes, including cellular differentiation (Nolte-'t Hoen et al, 2015), proliferation (Zhao et al, 2012), apoptosis (Yeh et al, 2012), angiogenesis (Caporali and Emanueli, 2011), the development of cancer , and the inhibition of tumor angiogenesis and growth (Luo et al, 2013;Tu et al, 2015). Endometriosis is a common, although mysterious and fascinating, gynecological condition with diverse clinical manifestations and high variability.…”

“…It has been reported that a class of endogenous, small non-coding miRNAs may act upon the mRNA of MAP4K4 and inhibit its translation (26). To further evaluate the regulating mechanisms of MAP4K4 in CRC, bioinformatics analysis was performed.…”

“…50 Additional clinical relevance is attributed to miR-30d as novel potential epigenetic drug for diabetes because it preserves mouse b-cells against TNFa-induced suppression of insulin expression and signaling. 51 Alternatively, the main target organs of insulin, namely liver, skeletal muscle, and adipose tissue might be also the targets of inflammatory pathways that could promote insulin resistance by means of epigenetic modifications (Fig. 1).…”
