MicroRNA-29a Is a Candidate Biomarker for Alzheimer’s Disease in Cell-Free Cerebrospinal Fluid

Abstract: The identification of reliable biomarkers for Alzheimer’s disease (AD) remains challenging. Recently, abnormal levels of microRNAs (miRNAs) miR-27a, miR-29a, miR-29b, and miR-125b in cerebrospinal fluid (CSF) of AD patients were reported. We aimed to confirm the biomarker potential of these miRNAs for AD diagnosis. Additionally, we examined the influence of blood contamination on CSF miRNA levels as potential confounding factor. We studied expression levels of the four miRNAs by quantitative PCR in CSF samples… Show more

“…For instance, up-regulated miR-34 is detected in AD-affected cells, which represses the degradation of tau and causes memory impairment through SIRT1 inhibition [284, 285]. Whereas, down-regulation of miR-29a in AD notably induces the expression of its downstream target BACE1 , which accelerates the cleavage from APP to Aβ [286, 287]. Moreover, several elevated miRNAs such as miR-9, miR-155, miR-146a stimulate neuro-inflammation around brain cells, which is a widely recognized mechanism for AD genesis [282].…”

“…Furthermore, miRNA levels in biofluids can be easily detected by standard molecular biology techniques such as quantitative PCR (qPCR). Hence, miRNAs are being considered as potential novel biomarkers in AD (24, 42, 43). …”
