MicroRNA-155 Suppresses Mesangial Cell Proliferation and TGF-β1 Production via Inhibiting CXCR5-ERK Signaling Pathway in Lupus Nephritis

Abstract: Increasing evidence shows miR-155 plays an important role in regulating inflammatory processes in systemic lupus erythematosus (SLE), especially in lupus nephritis (LN). Because the chemokine CXCL13 is implicated in the pathogenesis of LN, here we examined whether miR-155 can modulate the activity of CXCL13 or its receptor CXCR5. We determined the expression of CXCL13 in normal and MRL/lpr mice and found elevated levels of CXCL13 in the kidneys of MRL/lpr mice compared with normal kidneys. Besides, CXCL13 expr… Show more

“…ERK expression is increased in several immune diseases. 15,16 In our study, we showed that ACL titers significantly decreased under BRAF or MEK inhibitors. We cannot draw any conclusions about a specific effect of the MAPK pathway inhibition, or an anti-inflammatory effect on histiocytes.…”

“…The exact mechanism underlying miR-155-mediated SLE development remains unclear. Jie et al found that miR-155 inhibits the proliferation of mesangial cells and decreases the production of TGF-β1 in LN ( 24 ). Qian et al reported that miR-155 deficiency ameliorates autoimmune inflammation in SLE by targeting sphingosine-1-phosphate receptor 1 ( 25 ).…”

“…Krebs et al found that miR-155 stimulation in experimental crescentic glomerulonephritis resulted in accelerated kidney injury through T H 17 immunity [ 8 ]. However, another study reported that miR-155 decreased the proliferation of mesangial cells in lupus nephritis, which indicates the renoprotective characteristics of miR-155 [ 9 ]. Nevertheless, the actual relationship between miR-155 and renal disease may be much more complex.…”
