MicroRNA-133a Inhibits Osteosarcoma Cells Proliferation and Invasion via Targeting IGF-1R

Abstract: Background/Aims: MicroRNAs (miRNAs) are a class of small noncoding RNAs that regulate gene expression by repressing translation or cleaving RNA transcripts in a sequence-specific manner. Downregulated microRNAs and their roles in cancer development have attracted much attention. A growing body of evidence showed that microRNA-133a (miR-133a) has inhibitory effects on cell proliferation, migration, invasion, and metastasis of osteosarcoma. Methods: MiR-133a expression in human osteosarcoma cell lines and human … Show more

“…IGF1R likely performs its functions partly by participating in the regulation of AKT and ERK signalling pathways (26)(27)(28). To determine whether miR-495 is involved in the regulation of AKT and ERK pathways in HCC, we detected the expression levels of AKT, p-AKT, ERK and p-ERK in Hep3B cells transfected with miR-495 mimics or miR-NC.…”

“…For example, Bi et al (17) suggested that miR-100 serves as a cancer inhibitor in OS development via suppressing OS growth by regulating FGFR3 expression. MicroRNA-133a can prevent the proliferation and invasion of osteosarcoma cells by targeting IGF-1R (18). MiR-542-5p has been found to serve as an oncogene in OS though promoting OS proliferation by targeting HUWE1 (19).…”

“…U-2OS and SaOS-2 cells were cultured in McCoy’s 5A (Sigma, M4892) with 10% fetal bovine serum (CLARK, FB25015), MG-63 cells were cultured in MEM (Gibco 41500-034) with 10% fetal bovine serum, penicillin (100 U/mL) and streptomycin (100 U/mL) in a humidified incubator (Thermo Fisher, Runcorn, Cheshire, UK) with 5% CO 2 at 37°C [22, 23]. …”

“…At 24 hours after transfection, cells were treated with or without AG for additional 24 hours and collected for western blot. Transfected cells were also used for MTT, Immunocytochemistry, Live/dead assay, Wound healing assay, Migration and invasion assays [23]. …”
