MicroRNA-133a inhibits cell proliferation, colony formation ability, migration and invasion by targeting matrix metallopeptidase 9 in hepatocellular carcinoma

Abstract: Abstract. is downregulated in various types of human malignancy, including hepatocellular carcinoma (HCC), renal cell carcinoma, esophageal squamous cell carcinoma, bladder cancer, ileal carcinoid and rhabdomyosarcoma. The aim of the present study was to examine the effects of miR-133a on HCC cell proliferation, colony formation, migration and invasion. miR-133a was transfected into the HCC HepG2 and SMMC-7721 cell lines and the expression levels of miR-133a were determined; in addition, cell viability assays… Show more

“…And after knockdown of hsa_circ_0001649 with siRNA in immortalized hepatoma cell lines, HCC-LM3 and MHCC-97L (Fig. 4A), we tested the expression levels of several matrix metallopeptidases (MMPs) 9, 10, 2, 13, and 7, which had been reported to have key roles in promoting the metastasis of HCC [26][27][28][29]. We found significant increase in mRNA levels of MMP9, MMP10, and MMP13 (Fig.…”

“…miRNAs are believed to modulate more than a third of human genes (11) and therefore serve notable functions in a large number of biological processes, including cell proliferation, cell cycle, apoptosis, differentiation, metabolism, migration and invasion (12)(13)(14). Furthermore, increasing evidence has demonstrated that the abnormal expression of miRNAs contributes to the initiation and development of human malignancies, including HCC (15)(16)(17). Although numerous miRNAs have been identified, their expression and roles in HCC carcinogenesis and progression, and the underlying mechanisms by which this occurs, remain largely unidentified.…”

“…Therefore, understanding the molecular mechanisms underlying the carcinogenesis and progression of HCC is required for the development of novel therapeutic treatments for patients with HCC, in order to improve the rate of survival. A number of previous studies have demonstrated that abnormal expression levels of miRNAs may be important in HCC initiation and development, suggesting that miRNA may be investigated as a novel direction in the targeted therapy of HCC (26)(27)(28).…”
