MicroRNA-126 Overexpression Inhibits Proliferation and Invasion in Osteosarcoma Cells

Abstract: This study investigated the biological effects of microRNA-126 overexpression in human MG63 osteosarcoma cells. A recombinant plasmid expressing microRNA-126, pcDNA6.2-microRNA-126, was constructed and transfected into MG63 cells. Using real-time fluorogenic quantitative polymerase chain reaction, the microRNA-126 expression was measured in microRNA-126-MG63 group, Ctrl-MG63 group, and blank group. Cell proliferation, cell cycle distribution, cell migration, and invasion were analyzed using methyl thiazolyl te… Show more

“…Previous studies have shown that ADAM9 expressed on the cancer cell membrane participates in DDP resistance of clear cell carcinoma cells [17]. Strikingly, numerous studies revealed that ADAM9 acted as a miR-126 target, while miR-126 suppresses tumor cell proliferation, migration, and invasion by targeting ADAM9 [18][19][20][21][22][23][24][25]. Moreover, Caporali et al reported that miR-126-3p could inhibit growth and metastasis of dabalafini-resistant melanoma and enhance the drug sensitivity via regulating ADAM9 [26].…”

“…Previous studies also reported that miRNAs might target ADAM9 and contribute to diverse biological functions. For example, in osteosarcoma, miR-126 targeted ADAM9 to inhibit cell proliferation and invasion ( 40 ). In hepatocellular carcinoma, ADAM9 might be targeted by miR-1274a ( 41 ).…”

“…Our initial results revealed that miR-126 is significantly downregulated, while the mRNA and protein expression of EGFL7 and ERK are upregulated in HCC tissues. MiR-126 expression is downregulated in human cancers, reducing its ability to suppress the proliferative and invasive capacities of cancer cells, such as lung cancer, osteosarcoma, oral cancer and so on [ 11 , 12 , 21 ]. Wong et al have found that miR-126 is significantly decreased in patients carrying hepatitis B and C virus, and lower miR-126 expression predicts poor survival and tumor recurrence for HCC patients after surgery [ 22 ].…”
