SummaryAn efficient rapid and large-scale in vitro clonal propagation of the valuable medicinal herb Eclipta alba (Asteraceae) by enhanced axillary shoot proliferation in cotyledonary node segments was designed. The medium type, various carbon sources, plant growth regulators, and coconut water markedly influenced in vitro propagation of Eclipta alba. An in vitro plantlet production system has been investigated on Murashige and Skoog (MS) medium with the synergistic combination of benzyladenine (4.4 mM), kinetin (4.6 mM), 2-isopentenyladenine (4.9 mM), gibberellic acid (1.4 mM), 5% coconut water, and 3% sucrose which promoted the maximum number of shoots as well as beneficial shoot length. Subculturing of cotyledonary node segments on a similar medium enabled continuous production of healthy shoots with similar frequency. Rooting was highest (94.3%) on full strength MS medium containing 9.8 mM indolebutyric acid. Micropropagated plants established in garden soil, farmyard soil, and sand (2:1:1) were uniform and identical to the donor plant with respect to growth characteristics as well as floral features. These plants grew normally without showing any morphological variation.

A rapid and efficient plant propagation system through shoot tip explants was established in Vitex trifolia L., a medicinally important plant belonging to the family Verbenaceae. Multiple shoots were induced directly on Murashige and Skoog (MS) medium consisting of different cytokinins, 6-benzyladenine (BA), kinetin (Kin) and 2-isopentenyl adenine (2-iP), BA at an optimal concentration of 5.0 μM was most effective in inducing multiple shoots where 90 % explants responded with an average shoot number (4.4 ±0.1) and shoot length (2.0±0.1 cm) after 6 weeks of culture. Inclusion of NAA in the culture medium along with the optimum concentration of BA promoted a higher rate of shoot multiplication and length of the shoot, where 19.2±0.3 well-grown healthy shoots with an average shoot length of 4.4±0.1 cm were obtained on completion of 12 weeks culture period. Ex vitro rooting was achieved best directly in soilrite when basal portion of the shoots were treated with 500 μM indole-3-butyric acid for 15 min which was the most effective in inducing roots, as 95 % of the microshoots produced roots. Plantlets went through a hardening phase in a controlled plant growth chamber, prior to ex-vitro transfer. Micropropagated plants grew well, attained maturity and flowered with 92 % survival rate. The results of this study provide the first report on in vitro plant regeneration of Vitex trifolia L. using shoot tip explants.

An efficient protocol was developed for plant regeneration, multiplication and rooting under in vitro condition in pointed gourd. Highest percent of shoot regeneration was 91.66%, when nodal explants were cultured on MS+1.0 mg/1 BAP. The maximum number of shoots (4.8) per explant was observed in MS + 1.0 mg/1 BAP + 0.2 mg/1 NAA from nodal segment. Among the two explants, nodal segment was found better for shoot regeneration and multiplication. The best response towards root induction was achieved on half MS medium supplemented with 0.4 mg/1 NAA. The regenerated healthy rooted plantlets were transferred to small plastic pot containing garden soil and compost in a ratio of 2:1. Immediately after transplantation the plantlets were covered with polythene bag to prevent desiccation. After acclimatization the plantlets were successfully transferred to the garden soil and the percentage of survivability in such condition was 90%.
