Micropropagation of the Endangered and Decorative Specie Dianthus serotinus Waldst. et Kit.

Abstract: During past decades, great attention has been paid to propagation of endangered plant taxa in order to preserve biodiversity. The aim of this study was to optimize a protocol for in vitro propagation of the critically endangered and decorative species Dianthus serotinus Waldst. et Kit. The effects of different concentration of MS salt (Murashige and Skoog) of the culture, medium pH and different carbohydrates (sucrose, glucose, and fructose) on shoot multiplication were examined. The best results were obtained… Show more

“…In this study, a photoperiod of 16 h and full MS are the optimum conditions for production of nodes. Similar findings were observed on Dianthus serotinus; the number of nodes per shoot was In vitro seed germination and node culture of Grewia tenax reduced when seedlings were grown on half strength MS medium (Marković et al 2013). Variation in number of nodes according to media types is also reported (Chathurani et al 2009).…”

“…The concentration of MS salts (MS or half-strength MS) did not affect the frequency of shoot regeneration. On the other hand, there were some small differences in the percentage of regeneration depending on the explant type, since higher regeneration (Marković et al, 2013). The mean number of shoots regenerated per explant ranged between 2.6 and 4.6 (Table 2), but a statistically significant impact of a hormone or MS salt concentration on the number of shoots could not be observed.…”

“…The impact of the concentration of MS salts is not significant, but generally longer shoots develop on MS media. The impact of the concentration of MS salts on shoot length was recorded for D. serotinus and D. pinifolius (Marković et al, 2013(Marković et al, , 2016, with longer shoots developed on full strength MS media. Nevertheless, half strength MS media can have a favourable effect on shoot regeneration (D e s i l et s et al.…”

“…speciosus (Cristea et al, 2004, Holobiuc i Blindu, 2006, а код D. spiculifolius испитана је генетска стабилност биљака добијених in vitro, при чему је утврђено да је генетска варијабилност популације сачувана применом одговарајућег протокола микропропагације, док је сомаклонално варирање in vitro биљака минимално (Cristea et al, 2014). Код врсте D. serotinus је први пут код рода Dianthus испитано дејство различитих врста и концентрација шећера на развој култура у фази мултипликације, а с обзиром да расте на песку алкалне реакције, истражен је и утицај pH вредности медијума (Marković et al, 2013a).…”
