Biotechnology of the Banana: A Review of Recent Progress

A number of biotechnological tools have been developed which could help breeders to evolve new plant types to meet the demand of the food industry in the next century. Available techniques for the transfer of genes could significantly shorten the breeding procedures and overcome some of the agronomic and environmental problems which would otherwise not be possible through conventional methods. In vitro protocols have been standardized to allow commercially viable propagation of desired clones of Musa. An overview of the regeneration of banana by direct and indirect organogenesis, and somatic embryogenesis is presented in this article. In addition, the use of several other biotechnological techniques to enrich the genome of banana, such as selection of somaclonal variants, screening for various useful characteristics, cryopreservation, genetic transformation and molecular genetics are reviewed. In conclusion, the improvement of banana through modern biotechnology should help ensure food security by stabilizing production levels in sustainable cropping systems geared towards meeting domestic and export market demands.

Abstract:The study was undertaken to examine banana cv. Kanthali floral bud apex as an alternative source material for in vitro propagation because huge number of explants die due to microbial contamination in case of shoot tip explants. Contamination free cultures were established by treating the floral bud explants with 0.1% HgCl 2 for 6 min. This study found that inflorescence tissues of experimental plant was almost contamination free but was high in phenolic compounds. Phenolic compounds secretion was successfully stopped by pre-soaking them in an antioxidant solution of 0.125% potassium citrate: citrate. After antioxidant treatment the floral bud explants were cultured on MS medium supplemented with different concentrations and combinations of BA+Kn+IAA/IBA+15%CW. Compact, white/greenish white callus was formed in different amount at all concentrations after 3 weeks of culture. All were again subcultured at same medium and after another 30-35 days at 2.0 mg L 1 BA+2.0 mg L 1 Kn+2.0 mg L 1 IAA+15%CW some callus showed embryogenic structure.

The present investigation has been concentrated on the establishment of an efficacious method for in vitro regeneration of a Musa cultivar Gopi of Tripura. Focus has been greatly anchored to the sterilization procedure for assuring successful aseptic culture establishment of this cultivar. The culture protocol for shoot induction and proliferation comprised of four different treatments in basal MS medium fortified with 6-Benzylaminopurine of varying concentrations. The optimum response was encountered in the treatment of BAP in MS medium at concentration of 8mg l -1 for shoot bud induction and multiplication. Subsequent shoot growth and multiplication was achieved through repeated subcultures in media containing lower concentration (4 mg l -1 ). Root initiation was stimulated and brought about by treatments with two different concentrations of Indole butyric acid. The best rooting was manifested in treatment with IBA at concentration of 2mg l -1 . The present study is an approach towards successful establishment of a simple and rapid clonal propagation of a potent Musa cultivar Gopi having traditional and commercial value from this region of Tripura, North-east India.
