Micropropagation of a terrestrial Cymbidium species using rhizomes developed from seeds and pseudobulbs

Abstract: Growth of Cymbidium kanran rhizome was enhanced by higher NAA:BAP ratios in modified Murashige & Skoog (MS) media. Only vegetative shoots resulted from rhizomes cultured in vitro when lower NAA: BAP ratios were used. The rhizomes were induced from the axils of leaves when shoots were explanted to medium containing higher concentrations of NAA. Root formation of C. kanran was inhibited by the addition of either auxin or cytokinin to the culture media. Differentiation of the rhizomes into plantlets occurred when… Show more

“…The similar results were obtained in Cymbidium kanran (Shimasaki & Uemoto 1990) and Dendrobium aqueum (Philip et al, 2009). In Vanda coerulea the rooting efficiency varied with different concentrations of auxin such as IAA, IBA and NAA.…”

“…Paek and Kozai [18] reported 6-BA to be the best cytokinin for inducing shoot formation, for switching rhizome tissues into protocorm-like bodies (PLBs) and for direct formation of multiple shoots from branched rhizomes in most species of Cymbidium. Auxins (especially NAA) in combination with 6-BA are known to stimulate shoot initiation and proliferation from rhizome segments in Cymbidium kanran [19] and from pseudobulbs in T. latifolia [12], B. striata [20], P. hookeriana [11], P. bulbocodioides [21] and Cremastra appendiculata [22]. These results are similar to those of the present study on C. amoena.…”

“…Protocorms branch into rhizome-like structures during germination in species such as Cymbidium and Geodorum, a growth response which was enhanced with auxin application. 82,83 In addition, NAA and IAA helped to sustain rhizome growth in Spathoglottis, 84 and in Cymbidium 85 auxins induced rhizome growth while inhibiting shoot production. Further, researchers have reported auxin-stimulated production of rhizomes from seedling leaf axils, 8 and in adult plants, rhizogenesis was promoted by topical application of NAA on psuedobulbs of Cymbidium.…”
