Microplate assay for quantitation of neutral lipids in extracts from microalgae

Abstract: Lipid quantitation is widespread in the algae literature but popular methods including gravimetry, GC-MS, and Nile red cell staining suffer drawbacks including poor quantitation of neutral lipids, expensive equipment, and variable results among algae species, respectively. A high-throughput microplate assay was developed that utilizes Nile red dye to quantify neutral lipids that have been extracted from algae cells. Because the algal extracts contained pigments that quenched Nile red fluorescence, a mild bleac… Show more

“…The lipid content was determined by the Nile Red method as described elsewhere (Chen et al, 2009;Higgins et al, 2014). Commercial corn oil from a local store was used for the preparation of the calibration curve.…”

“…Lipid extracts were transesterified with 1 M methanolic HCl and analyzed by gas chromatography and mass spectrometry (GC‐MS) using methods previously described (Higgins et al, ). Briefly, internal standard (nonadecanoic acid) was added to lipid extracts that were then dried, re‐suspended in hexane, and transesterified.…”

“…In this study we used gas chromatography with mass spectrometry (GC‐MS), thin layer chromatography (TLC), and a neutral lipid assay (Higgins et al, ) to thoroughly describe the lipid content of axenic cultures and co‐cultures under autotrophic and mixotrophic conditions. Our objective was to determine the quantity of lipids conducive to biodiesel production and make inferences about potential biodiesel quality based on fatty acid data.…”

“…The resulting lipid extract was then used to carry out a neutral lipid assay [21] with slight modifications: specifically 1 μg/mL Nile Red solution was used rather than 0.5 μg/mL as discussed in the publication. Canola oil was used as a standard.…”
