Micropatterning gradients and controlling surface densities of photoactivatable biomolecules on self-assembled monolayers of oligo(ethylene glycol) alkanethiolates

Abstract: A photoimmobilization technique for decorating surfaces with micropatterns that consist of variable densities of bioactive molecules is described. The efficacy of the patterns for controlling cell adhesion and shape has been demonstrated. This technique is useful for the study of cell behavior on micropatterns.

“…Weber et al [183] grafted carbohydrates onto TiO 2 surfaces for applications in cell recognition, blood coagulation cascade and sialoglycoprotein recognition. Hypolite et al [184] and Herbert et al [185] synthesized bioconjugate molecules bearing both a peptide domain and photoactive group together with benzophenone. The benzophenone derivatives could be linked covalently onto the solid surface by irradiation.…”

“…Furthermore, fibronectin gradients generated by microfluidics induce fibroblast haptotaxis towards higher fibronectin concentration; however, the net movement directly correlates with the gradient slope, while the overall rate of migration does not (Rhoads and Guan, 2007). Herbert et al (1997) introduced a method for using photoimmobilization of peptides on self-assembled monolayer, while (Wang et al, 2004) merged electrochemical potential gradients with electrosorption of organothiols. Smith et al (2004) produced surface-bound gradients of fibronectin by the crossdiffusion of inert and reactive functionalized alkanethiols on gold.…”

“…CHO cells overexpressing EGF receptor proteins exhibited enhanced growth on higher density (smaller gap) of immobilized EGF. Herbert et al 66 photo-immobilized micropatterns of Arg-Gly Asp (RGD) sequencecontaining oligopeptides on an oligo(ethylene glycol) alkanethiolate SAM. The pattern and amount of immobilized RGD were controlled by manipulating the duration of UV exposure through a photomask, resulting in a surface density gradient of immobilized peptides.…”
