Microneedle array facilitates hepatic sinusoid construction in a large-scale liver-acinus-chip microsystem

Abstract: Hepatic sinusoids play a key role in maintaining high activities of liver cells in the hepatic acinus. However, the construction of hepatic sinusoids has always been a challenge for liver chips, especially for large-scale liver microsystems. Herein, we report an approach for the construction of hepatic sinusoids. In this approach, hepatic sinusoids are formed by demolding a self-developed microneedle array from a photocurable cell-loaded matrix in a large-scale liver-acinus-chip microsystem with a designed dua… Show more

“…Hepatocytes can be interfaced to the artificial blood vessel and this has been shown to improve hepatocyte function, as well as increasing sensitivity for drug toxicity testing (28). Perfusable liver sinusoids have also been developed by microneedleassisted fabrication enabling a dual blood supply, resulting in increased cell viability and hepatocyte metabolism (29). The authors also report the spontaneous emergence of secondary sinusoids, implying that in the right context, cells possess considerable degree of self-organization capacity.…”

“…Following Richard Feynman's groundbreaking 1959 discourse “There is Plenty of Room at the Bottom”, 14 continuing advancements in the conception and manufacture of active miniaturized components have prompted an array of explorations within the micro, nano, and quantum dimensions. 15,16 Nowadays, micro/nanorobots (MNRs) have extended our ability to observe, exploit and control the microscopic world. 17,18 MNRs refer to robotic agents with dimensions from the millimeter to nanometer range that are capable of transforming energy to movement and forces.…”

“…With the development of micromachining technology, organ chips have been employed for replicating various human diseases, benefiting from their advantages of small size, high degree of humanization, controlled microenvironments, and observable disease progression . Liver chips, as one of the extensively focused organ chips, , provide a promising alternative for modeling the evolution of colorectal cancer-induced thrombus . To date, liver chips have progressed from two-dimensional (2D) culture to three-dimensional (3D) culture .…”
