Micrometer-Sized Magnesium Whitlockite Crystals in Micropetrosis of Bisphosphonate-Exposed Human Alveolar Bone

Abstract: Osteocytes are contained within spaces called lacunae and play a central role in bone remodelling. Administered frequently to prevent osteoporotic fractures, antiresorptive agents such as bisphosphonates suppress osteocyte apoptosis and may be localized within osteocyte lacunae. Bisphosphonates also reduce osteoclast viability and thereby hinder the repair of damaged tissue. Osteocyte lacunae contribute to toughening mechanisms. Following osteocyte apoptosis, the lacunar space undergoes mineralization, termed … Show more

“…However, systemic driven HAP deposition can take place in the BrM, as reported before (Gorgels et al, 2012) (Alcolado et al, 1986). More recently, similarly structured spherules were also identified within patients with osteoporosis and in cardiovascular disease (Bertazzo et al, 2013;Shah et al, 2017). Thus, comparable mineralization mechanisms in a variety of non-osseous tissues appear to be associated with a number of different disease conditions.…”

“…Osteocytes are believed to be able to detect microdamage through ruptured osteocyte processes . Moreover, osteocyte lacunae (Ot.Lc) contribute toward cortical toughening through crack deflection, while occlusion of Ot.Lc by mineral deposition gives rise to isolated regions of lower strain‐to‐failure than the extracellular matrix . However, similar osteocyte densities as well as similar size and shape distributions (i.e., similar occupied area fraction) for all of the experimental groups suggests comparable biomechanical capacities in 16‐ and 20‐week‐old OVX and Ctrl animals.…”

“…The collagen crosslink ratio, taken as the % area of the 1662 cm −1 sub-component, corresponds to biochemically determined pyridinoline content and represents pyridinoline trivalent crosslinks-one of the several naturally occurring crosslinks in collagen [30]. Here, the collagen crosslink Phe levels decline with increasing tissue maturity [48] and are typically recorded at around 9-12% with respect to the intensity of the phosphate peak at 960 cm −1 [49,50] in periimplant bone. Here, Phe levels for Smokers (12-32%) and Ctrl (8-33%) were relatively higher, which reflects the contribution of rapidly formed woven bone.…”
