Microgravity-Induced Alterations of Inflammation-Related Mechanotransduction in Endothelial Cells on Board SJ-10 Satellite

Abstract: Endothelial cells (ECs) are mechanosensitive cells undergoing morphological and functional changes in space. Ground-based study has provided a body of evidences about how ECs can respond to the effect of simulated microgravity, however, these results need to be confirmed by spaceflight experiments in real microgravity. In this work, we cultured EA.hy926 ECs on board the SJ-10 Recoverable Scientific Satellite for 3 and 10 days, and analyzed the effects of space microgravity on the ECs. Space microgravity suppre… Show more

“…Aside from this, changes in gravity affect the organization of cytoskeletal proteins which orchestrate adaptation processes to mechanical stress 1618 . Thus, permanent remodelling of the cytoskeleton by alternating µg and hyper-g conditions has also an effect on cell mechanical stability 19,20 , motility and adhesion 21 and cell signalling 11 .…”

“…This device was mounted in the SJ-10 satellite launched on April 6 and recovered on April 18, 2016 (Supplemental Fig. S1C) (23,24). At 36 h before launch, 2 cell culture chambers with an effective culture area of 12 cm 2 each were filled with a cell suspension containing 5 3 10 5 rBMSCs per chamber and placed in an incubator for 12 h at 37°C to allow cell attachment.…”

“…Each substrate was then cut into pieces and used for scanning electron microscope (SEM) imaging or immunofluorescence (IF) staining. The SEM imaging was conducted as previously described (24). In short, 1 aliquot of the samples was dehydrated with ethanol from 50-70-80-90-100% for 5 min at each gradient, transferred to tert-butyl alcohol twice, dried at the critical point, coated with a thin layer of gold, and finally imaged by an FEI Quanta 200 SEM (Thermo Fisher Scientific).…”
