Microglia Development and Maturation and Its Implications for Induction of Microglia-Like Cells from Human iPSCs

Abstract: Microglia are resident immune cells of the central nervous system and play critical roles during the development, homeostasis, and pathologies of the brain. Originated from yolk sac erythromyeloid progenitors, microglia immigrate into the embryonic brain parenchyma to undergo final postnatal differentiation and maturation driven by distinct chemokines, cytokines, and growth factors. Among them, TGFβ1 is an important regulator of microglial functions, mediating homeostasis, anti-inflammation, and triggering the… Show more

“…These cells can be differentiated into neurons to investigate not only neurogenesis but also Aβ production. It would be also possible to generate in vitro models of the BBB [ 91 ] or microglial cells [ 92 ] to further study Aβ clearance from CNS, and therefore to gain insight into the molecular role of ABCA7 in AD.…”

“…In this paper, we investigated the possible therapeutic efficacy of the adoptive transfer of microglia, modified with the rAAV vector carrying IL-15, to mimic the EE effects on glioma. The possibility to exploit microglial cells intranasally delivered as a source of IL-15 directly in the tumor mass could be an attractive alternative for GBM patient treatment, due to the high potential for clinical translatability, as one can obtain the microglia starting from patients' iPSCs, as recently reported (46). The local delivery of cells as therapeutic carriers against glioma has produced encouraging results but encounters obstacles with regard to the repeatability and invasiveness of administration.…”

“…Genetic data has been invaluable in providing a greater understanding of microglia and their function in neurological conditions. Being derived from the early erythro-myeloid progenitors in the yolk sac ( Ginhoux et al, 2010 ), many genes contribute to their differentiation and role as immune cells within the CNS ( Wurm et al, 2021 ). As aging occurs, microglial gene expression changes and this might potentially explain why aging is the major contributor to disease risk in neurodegenerative conditions ( Olah et al, 2020 ).…”
