Microfluidic tool based on the antibody‐modified paramagnetic particles for detection of 8‐hydroxy‐2′‐deoxyguanosine in urine of prostate cancer patients

Abstract: Guanosine derivatives are important for diagnosis of oxidative DNA damage including 8-hydroxy-2'-deoxyguanosine (8-OHdG) as one of the most abundant products of DNA oxidation. This compound is commonly determined in urine, which makes 8-OHdG a good non-invasive marker of oxidation stress. In this study, we optimized and tested the isolation of 8-OHdG from biological matrix by using paramagnetic particles with an antibody-modified surface. 8-OHdG was determined using 1-naphthol generated by alkaline phosphatase… Show more

“…Cytosine-adenine reduction (CA peak) was detected by square wave voltammetry coupled with adsorptive transfer technique (AdTS SWV) [45,46,47,48,49,50]. The parameters of the electrochemical determination were as it follows: initial potential 0 V; end potential –1.85 V; frequency 80 Hz; potential step 0.005 V; amplitude 0.025 V. Time of accumulation was optimized.…”

“…denaturation [107]. Indirect detection with 1-naphthol generated after 8-OHdG immunoseparation using paramagnetic particles from biological sample has successfully been implemented [40] and it is shown in Fig. Nanoparticles have a lot of physico-chemical advantages [109,110].…”

“…This includes 8-hydroxy-2′-deoxyguanosine (8OHdG) which acts as an indicator of DNA oxidation (Ogino and Wang, 2007;Valavanidis et al, 2009;Zwart et al, 1999). Nowadays, the methods used to quantify this biomarker involve complex, expensive and non-portable techniques (Fan et al, 2012;Li et al, 2011;Wang et al, 2011;Zitka et al, 2011), which impair a more effective utilization of OS biomarkers as a predictive tool for degenerative disease installation. Only an immunosensor is presented very recently in literature for 8OHdG, but the overall procedure is very complex (Darwish et al, 2013).…”
