A molecularly imprinted polymer (MIP) was synthesized and evaluated to selectively extract ibuprofen, a nonsteroidal anti-inflammatory drug (NSAID), from human urine. The synthesis of the MIP was performed in dimethylformamide with acrylamide as functional monomer, ethylene glycol dimethacrylate as the cross linker and ibuprofen template as the target drug. The performance of the synthesized MIP as solid phase extraction (SPE) packing to recognize and bind ibuprofen was evaluated against other structurally related NSAIDs such as naproxen and ketoprofen. Using a mixture of acetonitrile-water (1/9 v/v) as a solvent for ibuprofen the binding recovery at approximately 90% was obtained for developed MIP in optimized conditions. Batch rebinding capacity of ibuprofen was determined from the derived Freundlich isotherm and was found to be 1.45 mol g -1 . Furthermore, a higher selectivity of developed MIP for ibuprofen over structurally related analogues was observed. The synthesized MIP has enabled the direct percolation of humane urine and the easy elimination of endogenous compounds from it with simple aqueous washing of the MIP-SPE packing. HPLC analysis has confirmed the high extraction recovery (ca. 85 %) of ibuprofen from such urine samples with use of proposed MIP-SPE system.

This review article deals with molecularly imprinted polymers (MIPs) as affinity-based separation media for sample preparation. An over view of two types of MIPs (molecularly imprinted particle and monolith) used for the sample preparation and modes of molecularly imprinted SPE (online mode, offline mode, on-column extraction, SPME, and microextraction in packed syringe) is given, focusing on the advantages and disadvantages of these types and modes. Next, problems (template leakage and incompatibility with aqueous conditions) associated with molecularly imprinted SPE and how to overcome those problems are described. Finally, pharmaceutical, food, bioanalytical, and environmental application of molecularly imprinted SPE will be discussed.

For the first time, a molecularly imprinted polymer (MIP) was synthesized by a noncovalent imprinting approach for the selective extraction of an illicit drug, LSD, from hair and urine samples. For the synthesis of MIP, an analog of LSD, was taken as a dummy template, methacrylic acid as a functional monomer, and ACN as a porogen solvent. The MIP was used for offline extraction before HPLC-MS analysis. By studying the interactions taking place between the LSD and the MIP, a selective procedure was established in organic media and applied to hair samples. By this way, 0.1 ng/mg of LSD was successfully detected in hair with 82% of extraction recovery. A low retention was also obtained on the control polymer (only 9%). This procedure was then modified to obtain a selective extraction in aqueous media for the determination of LSD in urine samples. The comparison with a conventional C18 clearly demonstrated the selectivity brought by the MIP to the determination of LSD in urine. LSD was easily detected in urine at only 0.5 ng/mL with 83% of extraction recovery on the MIP and 11% on the NIP. An LOQ of 0.2 pg/mL was estimated in urine samples.
