Microchip Emitter for Solid-Phase Extraction–Gradient Elution–Mass Spectrometry

Abstract: A microchip electrospray emitter with a magnetic bead trap has been designed for solid-phase extraction-gradient elution-mass spectrometry (SPE-GEMS). The goal of this method is the detection of analytes at low concentrations and it is here demonstrated using reverse phase coated magnetic beads (Mbs) for the preconcentration and detection of the peptides. The sample is passed through the chip, and the peptides are retained and enriched in the trap. After washing, the peptides are released sequentially by stepw… Show more

“…After preconcentration, analytes can be recovered for further processing or detected directly. Gasilova et al 15 used C18 functionalized magnetic beads integrated into a microfluidic device coupled to ESI-MS for peptide preconcentration. Microfluidic devices integrating a functionalized solid phase are promising devices for digestion, preconcentration or separation: Ethier et al 16 used SCX solid phase to make a proteomic reactor that allowed protein extraction, clean-up, preconcentration, and sample digestion.…”

“…Strategy C produced the highest number of plasma protein groups and newly identified proteins among all the strategies tested. An SPE‐gradient elution‐MS microchip with a RP‐coated magnetic bead trap was designed for the detection and analysis of peptides of low concentration . The magnetic bead containing C8 and C18 chromatographic coatings trapped by permanent magnets was used as the SPE sorbent.…”
