Microcalorimetric Determination of the Effects of Amoxicillin, Metronidazole, and Their Combination on In Vitro Biofilm

Abstract: IMC gives new evidence about antibiotic effects on oral biofilms and is more informative than conventional culture and live/dead assays. The combination of antibiotics was found to be more efficient than metronidazole alone; however, only minor differences in growth inhibition were detected compared with amoxicillin alone.

“…In addition, there are now a rather large number of studies that have shown that microcalorimetric data could be used directly to monitor bacterial growth. 3,[16][17][18][19] Especially, flow-through systems have been of great interest in study biofilm metabolisms and eradication. 34,35 Finally, one should not restrict the use of IMC to monitoring the growth on surface but also within porous material such as artificial HA bone grafts for example.…”

“…Bacterial growth at the surface of the coating or in the pores of an open-porous scaffold can then be monitored and analyzed using the heat dissipated through the bacterial metabolism. 3,[16][17][18][19] As only heat production is measured by a peltier element placed between the sample container and the heat sink, solid sample can be analyzed easily. Indeed, previous studies have shown that growth and antimicrobial activity could be determined accurately in solid medium using IMC 20,21 or gels.…”

“…Astasov-Frauenhoffer et al were able to prove complete growth-inhibiting effects of amoxicillin and metronidazole on Streptococcus sanguinis , Porphyromonas gingivalis and Fusobacterum nucleatum apart from each other, but the combination was found to be more efficient than metronidazole alone [72]. Comparing local antibiotic therapy with photodynamic therapy, Bassetti et al presented no differences in reduction of pocket depths or reduction of the number of bacteria in the periodontal pockets [73].…”

“…For instance, Clauss et al (2013) monitored biofilms of S. aureus growing on human and bovine bone grafts, while Astasov-Frauenhoffer et al (2012) investigated the variability and dynamics of a triple-species biofilm and determined the efficacy of amoxicillin and metronidazole combinations against biofilms of various species (Astasov-Frauenhoffer et al 2014). Said et al (2014a) determined the efficacy and mechanism of action of an antibiofilm wound dressing.…”
