Microbial succession in an inflated lunar/Mars analog habitat during a 30-day human occupation

Abstract: BackgroundFor potential future human missions to the Moon or Mars and sustained presence in the International Space Station, a safe enclosed habitat environment for astronauts is required. Potential microbial contamination of closed habitats presents a risk for crewmembers due to reduced human immune response during long-term confinement. To make future habitat designs safer for crewmembers, lessons learned from characterizing analogous habitats is very critical. One of the key issues is that how human presenc… Show more

“…These environments are typified by enclosures associated with extraterrestrial habitation. The National Aeronautics and Space Administration (NASA) has a strong interest and motivation to understand the microbial content and ecology of these environments, particularly the International Space Station (ISS) as a test bed for other analogs of closed systems [5] and future human habitation on Mars [6]. …”

“…All the BiSKits were prepared following the procedure described elsewhere [7, 51]. Briefly, the sterile phosphate buffer saline (PBS) provided by the distributor was discarded from the bottle and replaced with 15 mL of sterile UltraPure DNA free PBS (MoBio Laboratories Inc, Carlsbad, CA).…”

“…The bacterial and archaeal microbiome succession of the ILMAH using NGS has been carried out [7]. This is the first report characterizing the succession of fungi in a simulated closed system meant for human habitation on other planets utilizing both traditional and state-of-the-art molecular techniques.…”

“…Hence, one of the major challenges in manned space exploration is protecting astronauts from microbiota dysbiosis. In recent years, with the development of high-throughput sequencing technology for complex microbial communities, both hostderived and environmental microbiome have frequently been studied, either in space stations (Checinska et al, 2015;Moissleichinger et al, 2016;Mora et al, 2016b;Be et al, 2017) or ground-based space simulations Mayer et al, 2016;Sun et al, 2016;Blachowicz et al, 2017;Schwendner et al, 2017;Turroni et al, 2017).…”
